Abstract

Kunyit merupakan tanaman herbal yang banyak dibutuhkan oleh industri obat tradisional. Penyakit bercak daun merupakan salah satu penyakit utama yang menyerang tanaman kunyit di Indonesia. Penyakit ini yang disebabkan oleh jamur Colletotrichum capsici. Pengendalian biologi dengan memanfaatkan agens hayati yang bersifat antagonis. Agens hayati seperti jamur endofit dapat ditemukan di dalam jaringan tanaman. Tujuan penelitian ini untuk mengkaji potensi jamur endofit yang ditemukan sebagai agens antagonis untuk menekan pertumbuhan C. capsici penyebab penyakit bercak daun. Penelitian dilakukan di Laboratorium Penyakit Tumbuhan, Jurusan Hama dan Penyakit Tumbuhan, Fakultas Pertanian, Universitas Brawijaya mulai bulan Januari 2019 sampai dengan Juni 2019. Penelitian terdiri dari beberapa tahapan: 1) isolasi jamur patogen C. capsici, 2) uji Postulat Koch, 3) isolasi jamur endofit tanaman kunyit, 4) uji patogenisitas jamur endofit, 5) uji penghambatan jamur endofit terhadap jamur patogen C. capsici. Hasil isolasi jamur patogen menunjukkan kenampakan makroskopis dan mikroskopis yang sama dengan patogen C. capsici. Hasil isolasi jamur endofit diperoleh total 15 isolat. Setelah dilakukan uji patogenisitas, terdapat 10 isolat yang tidak bersifat patogenik dengan ditandai tidak terdapat gejala nekrosis pada daun kunyit. Kemudian, jamur endofit yang tidak bersifat patogenik, dilakukan pengujian antagonis dengan jamur patogen C. capsici. Dari hasil pengujian antagonis, terdapat 9 jamur endofit yang memiliki potensi sebagai agen antagonis terhadap patogen C. capsici. Hasil uji antagonis menghasilkan presentase daya hambat yang berbeda antar perlakuan. Persentase tersebut diperoleh dengan menghitung daya hambat patogen C. capsici selama 7 hari. Sembilan jamur endofit yang telah diuji antagonis mampu menghambat pertumbuhan jamur patogen C. capsici. Dengan persentase penghambatan berkisar antara 10,72% sampai 58,80%.

English Abstract

Turmeric is an herbal plant that is much needed by the traditional medicine industry. Leaf spot disease is one of the main diseases affecting turmeric plants in Indonesia. This disease is caused by the fungus Colletotrichum capsici. Biological control by utilizing biological agents that are antagonistic. Biological agents such as endophytic fungi can be found in plant tissues. The purpose of this study was to examine the potential of endophytic fungi found as antagonistic agents to suppress the growth of C. capsici causing leaf spot disease. The research was conducted at the Laboratory of Plant Diseases, Department of Plant Pests and Diseases, Faculty of Agriculture, University of Brawijaya starting in January 2019 until June 2019. The study consisted of several stages: 1) isolation of pathogenic fungi C. capsici, 2) Postulate Koch test, 3) isolation of turmeric plant endophytic fungi, 4) endophytic fungal pathogenicity test, 5) endophytic fungal inhibition test on fungal pathogen C. capsici. The results of isolation of pathogenic fungi showed macroscopic and microscopic appearance similar to pathogen C. capsici. The results of isolation endophytic fungi obtained a total of 15 isolates. After a pathogenicity test, 10 isolates which were not pathogenic were marked with no symptoms of necrosis on turmeric leaves. Then, endophytic fungi that are not pathogenic, doing antagonistic testing with C. capsici fungal pathogen. From the results of antagonistic testing, there are 9 endophytic fungus that have the potential as antagonistic agents against C. capsici. The results of antagonistic tests produced a different percentage of inhibition between treatments. This percentage is obtained by calculating the inhibitory pathogen of C. capsici for 7 days. Nine endophytic fungus that have been tested by antagonists are able to inhibit the growth of the pathogen fungi C. capsici, with the percentage of inhibition ranging from 10.72% to 58.80%.
