Abstract
Tanaman lidah buaya mulai banyak dibudidayakan karena memiliki manfaat yang baik antara lain dalam bidang kesehatan, kosmetik dan pangan. Namun dalam budidaya lidah buaya yang dikembangkan terdapat permasalahan salah satunya adanya serangan penyakit busuk lunak yang disebabkan oleh bakteri Erwinia sp. Setiap tanaman memiliki ketahanan masing-masing dalam menghambat pertumbuhan patogen yang dibantu oleh mikroorganisme salah satunya bakteri. Bakteri yang berada dalam jaringan tanaman tanpa menyebabkan kerusakan disebut bakteri endofit. Tetapi sampai saat ini informasi mengenai bakteri endofit lidah buaya masih terbatas. Penelitian ini bertujuan untuk mengkaji keefektifan bakteri endofit tanaman lidah buaya dalam menghambat patogen Erwinia sp. dan mengetahui karakteristik bakteri tersebut Penelitian dilaksanakan di Laboratorium Penyakit Tumbuhan Jurusan Hama dan Penyakit Tumbuhan Fakultas Pertanian Universitas Brawijaya Malang pada bulan Desember 2018 – April 2019. Tahapan penelitian yaitu isolasi bakteri patogen Erwinia sp. penyebab busuk lunak lidah buaya, isolasi bakteri endofit pada bagian akar dan batang lidah buaya, seleksi, uji hipersensitif bakteri endofit, uji antagonis, dan karakterisasi bakteri sampai tingkat genus. Berdasarkan hasil isolasi patogen dari tanaman lidah buaya bergejala busuk lunak diperoleh sebanyak 12 isolat bakteri. Dari pengujian karakterisasi, identifikasi dan patogenesitas ditemukan 2 isolat bakteri tergolong kedalam genus Erwinia sp. yang diperkirakan sebagai patogen Erwinia chrysanthemi penyebab busuk lunak tanaman lidah buaya. Hasil isolasi bakteri endofit pada tanaman lidah buaya didapatkan sebanyak 86 isolat bakteri yang terdiri dari 48 isolat dari bagian akar dan 38 isolat dari bagian batang. Seleksi bakteri endofit yang memiliki kemampuan dalam menghambat patogen Erwinia sp. penyebab busuk lunak sebanyak 39 isolat. Enam isolat dengan penghambatan tertinggi dipilih untuk digunakan dalam uji antagonis lanjutan yaitu isolat B3.3C, A1.4B, B3.3B, A5.2A, A.1.1B dan B1.5A. Pengujian antagonis yang dilakukan didapatkan hasil bahwa keenam isolat bakteri endofit memiliki kemampuan penghambatan yang lebih baik dibanding bakterisida Streptomisin dengan rerata penghambatan tertinggi ditunjukkan pada isolat A5.2A, B3.3C dan B3.3B. Identifikasi dan karakterisasi pada bakteri antagonis yang memiliki kemampuan dalam menghambat patogen Erwinia sp. didapatkan hasil sebanyak 5 isolat tergolong kedalam genus Pantoea sp. yaitu isolat B3.3C, B3.3B, A1.4B, A1.1B dan B1.5A serta 1 isolat tergolong kedalam genus Erwinia sp. yaitu isolat A5.2A
English Abstract
Aloe vera plants are widely cultivated because they have good benefits in the fields of health, cosmetics and food. But in the development of aloe vera, there were problems, one of which was the attack of soft rot caused by the bacteria Erwinia sp. Each plant has its own resistance in inhibiting the growth of pathogens assisted by microorganisms, one of which is bacteria. Bacteria that are in plant tissues without causing damage are called endophytic bacteria. But until now information about aloe vera endophytic bacteria is still limited. This study aims to examine the effectiveness of aloe vera endophytic bacteria in inhibiting pathogen Erwinia sp. and know the characteristics of these bacteria The research was conducted at the Laboratory of Plant Diseases Department of Plant Pests and Diseases of the Faculty of Agriculture, Brawijaya University Malang in December 2018 - April 2019. The stages of the research were the isolation of pathogenic bacteria Erwinia sp. the causes of soft aloe vera, isolation of endophytic bacteria in the roots and aloe stem, selection, hypersensitivity test of endophytic bacteria, antagonistic test, and bacterial characterization to the genus level. Based on the results of isolation of pathogens from soft rot of the aloe vera plant obtained 12 bacterial isolates. From characterization, identification and pathogenicity tests found 2 isolates belonging to the genus Erwinia sp. it is thought that the pathogen Erwinia chrysanthemi causes soft rot of the aloe vera plant. The result isolation of endophytic bacteria in aloe vera plants was obtained as many as 86 bacterial isolates consisting of 48 isolates from the root section and 38 isolates from the stem section. Selection of endophytic bacteria that have the ability to inhibit pathogens Erwinia sp. causes of soft rot as many as 39 isolates. Six isolates with the highest inhibition were selected for use in the advanced antagonist test, namely isolates B3.3C, A1.4B, B3.3B, A5.2A, A.1.1B and B1.5A. The antagonistic test conducted showed that the six endophytic bacterial isolates had better inhibitory ability than bactericidal Streptomycin with the highest inhibition rate shown in isolates A5.2A, B3.3C and B3.3B. Identification and characterization of antagonistic bacteria that have the ability to inhibit pathogens Erwinia sp. the results obtained as many as 5 isolates belonging to the genus Pantoea sp. there are isolates B3.3C, B3.3B, A1.4B, A1.1B and B1.5A and 1 isolate belonging to the genus Erwinia sp. namely A5.2A isolates
