Abstract

Kentang (Solanum tuberosum L.) merupakan salah satu komoditas potensial sebagai sumber karbohidrat serta memiliki arti penting pada perekonomian dunia terutama di Indonesia. Salah satu kendala produksi kentang adalah serangan penyakit busuk lunak yang disebabkan oleh bakteri Erwinia sp., baik ketika masih di lapangan maupun di gudang penyimpanan. Penyakit tersebut dapat diatasi dengan melakukan pengendalian yang ramah lingkungan dan berkelanjutan yaitu dengan menggunakan varietas yang tahan terhadap serangan Erwinia sp. dan memanfaatkan agens hayati sebagai biokontrol patogen penyebab penyakit tumbuhan. Pada penelitian sebelumnya telah ditemukan beberapa genus bakteri non patogen pada rizosfer di UB Forest. Penelitian bertujuan untuk mengetahui potensi bakteri rizosfer di UB Forest untuk mengendalikan dan menghambat patogen Erwinia sp. penyebab penyakit busuk lunak pada tiga varietas umbi kentang. Penelitian dilaksanakan di Laboratorium Penyakit Tumbuhan, Jurusan Hama dan Penyakit Tumbuhan, Fakultas Pertanian, Universitas Brawijaya mulai Bulan Februari sampai dengan Mei 2019. Penelitian terdiri dari beberapa tahapan: (1) Perbanyakan inokulum bakteri patogen dan bakteri rizosfer di UB Forest (2) Uji patogenesitas bakteri patogen Erwinia sp. (3) Seleksi bakteri rizosfer terhadap patogen Erwinia sp. (4) Pengujian antagonis bakteri rizosfer terpilih terhadap patogen Erwinia sp. secara in vitro (5) Uji antibiosis bakteri antagonis (6) Uji penghambatan perkembangan penyakit busuk lunak oleh bakteri rizosfer pada tiga varietas Umbi Kentang yaitu Granola Kembang, Desiree, dan Atlantik. Berdasarkan hasil seleksi 30 bakteri rizosfer, terdapat 14 isolat bakteri rizosfer UB Forest yang bersifat antagonis terhadap patogen Erwinia sp. dan empat isolat memiliki diameter penghambatan tertinggi yaitu isolat Bacillus sp. (N3), Clostridium sp. (N10), Pseudomonas sp. (N17) dan Pseudomonas sp. (N19). Empat isolat bakteri antagonis dengan penghambatan tertinggi mekanisme antibiosisnya bersifat bakteriostatik dan hanya dapat menghambat pertumbuhan patogen Erwinia sp. Berdasarkan hasil analisis sidik ragam, interaksi perlakuan jenis isolat dan jenis varietas berpengaruh nyata terhadap massa busuk lunak umbi kentang. Pada perlakuan isolat bakteri antagonis Pseudomonas sp. (N19) pada varietas Desiree (V2B4) menghasilkan massa busuk lunak yang lebih rendah daripada kombinasi perlakuan yang lain, namun rerata massa busuk lunak tersebut masih lebih tinggi apabila dibandingkan perlakuan kontrol positif bakterisida pada varietas Desiree (V2B5). Persentase penurunan berat umbi terendah terdapat pada seluruh perlakuan isolat bakteri antagonis pada varietas Granola dan varietas Desiree dengan persentase 3%. Pada persentase penunuran berat umbi tertinggi terdapat pada perlakuan Clostridium sp. pada varietas Atlantik (V3B2) dengan persentase 5%.

English Abstract

Potato plant (Solanum tuberosum L.) is one of the potential commodities as a source of carbohydrates and it has an important meaning in the world economy, especially in Indonesia. One of the obstacles in potato production is the attack of soft rot disease caused by Erwinia sp. while in the field or in the storage warehouse. The disease can be overcome by carrying out environmentally friendly and sustainable controls by using varieties which are resistant to the attacks of Erwinia sp. and utilizing biological agents as biocontrol pathogens that cause plant diseases. In previous studies, several genera of non-pathogenic bacteria have been found in the rhizosphere of UB Forest. This study was conducted to determine the potential of rhizosphere bacteria in UB Forest against the pathogen Erwinia sp. causes of soft rot in three potato tuber varieties. The research was conducted at the Laboratory of Plant Diseases, Department of Plant Pests and Diseases, Faculty of Agriculture, Universitas Brawijaya from February to May 2019. The study consisted of several stages: (1) Propagation of pathogenic bacteria and rhizosphere bacteria inoculars in UB Forest. (2) Pathogenicity test of pathogenic bacteria Erwinia sp. (3) Selection of antagonistic of rhizobacteria from UB Forest to Erwinia sp. (4) Testing of selected rhizosphere bacterial antagonists to the pathogen Erwinia sp. in vitro (5) Antibiosis test for antagonistic bacterial (6) Test for inhibition of the development of soft rot by rhizosphere bacteria in three varieties of potato tuber. Based on the selection of 30 rhizosphere bacteria, there are 14 isolates of rhizosphere bacteria that are antagonistic to the pathogen Erwinia sp. and four isolates had the highest inhibitory diameter, namely Bacillus sp. (N3), Clostridium sp. (N10), Pseudomonas sp. (N17) and Pseudomonas sp. (N19). The four isolates of antagonistic bacteria with the highest inhibition of their antibiotic mechanism are bacteriostatic and can only inhibit the growth of the pathogen Erwinia sp. Based on the analysis of variance, the interaction of the treatment of isolates and varieties significantly affected the soft rot mass of potato tubers. In the treatment of Pseudomonas sp. (N19) in Desiree varieties (V2B4) produced lower soft rot mass compared to other treatment combinations, but the mean soft rot mass was still higher than the bactericidal control treatment in Desiree varieties (V2B5). The lowest percentage of tuber weight reduction was found in all antagonistic bacterial isolate treatments on Granola and Desiree varieties with a percentage of 3%. The highest percentage of tuber weight reduction is found in the treatment of Clostridium sp. in Atlantic varieties (V3B2) with a percentage of 5%.
