Abstract
Kanker mammae merupakan tumor ganas yang dapat tumbuh di dalam jaringan payudara yang dapat dipicu oleh agen karsinogenik DMBA (Dymethylbenz(α) Anthrasen). Kanker mammae akibat induksi DMBA mempunyai kemungkinan untuk menyebar ke organ lain seperti uterus. ROS akibat kanker mammae akan terbawa aliran darah menuju uterus. Peningkatan ROS dan inflamasi sistemik pada organ reproduksi seperti uterus yang ditandai dengan peningkatan ekspresi IL-1α dan TNF-α. Tujuan penelitian ini adalah untuk mengetahui efek terapi kombinasi Kurkumin dengan Vitamin E dalam menurunkan ekspresi IL-1α dan TNF-α tikus model kanker mammae yang diinduksi oleh MLD-DMBA pada organ uterus. Pembuatan hewan model kanker mammae dilakukan dengan induksi DMBA dosis 10 mg/kg BB dan estrogen dosis 20.000 IU/kg BB. Penelitian ini dibagi dalam 5 kelompok perlakuan yaitu kelompok kontrol negatif (KN), kontrol positif (KP), perlakuan satu (P1) terapi dengan Kurkumin 48mg/kg BB dan Vitamin E 300 IU/ekor, perlakuan dua (P2) dosis 72mg/kg BB dan 200 IU/ekor, dan perlakuan tiga (P3) dosis 108mg/kg BB dan 100 IU/ekor. Ekspresi IL-1α dan TNF-α diamati dengan Imunohistokimia. Analisis data dilakukan dengan uji ANOVA dan jika terdapat perbedaan nyata hasil antar perlakuan, maka dilakukan uji Tukey. Hasil penelitian menunjukkan bahwa kombinasi terapi kurkumin dan vitamin E mampu menurunkan ekspresi IL-1α dan ekspresi TNF-α secara signifikan (p<0,05) dengan dosis terapi 2 (kurkumin 72mg/kg BB + vitamin E 200 IU/ekor) yang paling efektif dalam menurunkan ekspresi IL-1α sebesar (5,76 ± 0,43%) dan menurunkan ekspresi TNF-α sebesar (6,21 ± 0,67%). Kesimpulan dari penelitian ini adalah kombinasi kurkumin dan vitamin E dapat digunakan sebagai terapi kanker mammae
English Abstract
Breast cancer is a malignant tumor that can grow in the breast tissue that can be triggered by the carcinogenic agent DMBA (Dymethylbenz (α) Anthrasen). DMBA-induced mammary cancer have the possibility to spread to other organs such as the uterus. ROS as a result of breast cancer will be carried by the blood flow to the uterus. Increased ROS and systemic inflammation of the reproductive organs such as the uterus characterized by increased expressionIL-1α and TNF-α, The purpose of this study was to determine the effects of curcumin combination therapy with Vitamin E in reducing the expression of IL-1α and TNF-α mouse models of breast cancer induced by MLD-DMBA in the organs of the uterus. Manufacture of animal models of breast cancer conducted by DMBA induced a dose of 10 mg / kg and estrogen dose 20,000 IU / kg. The research was divided into 5 groups: control group a negative (KN), a positive control (KP), the treatment of one (P1) treatment with curcumin 48mg / kg and Vitamin E 300 IU, treatment of two (P2) a dose of 72mg / kg BB and 200 IU, and treatment three (P3) dose of 108mg / kg and 100 IU. Expression IL-1α and TNF-α Immunohistochemistry observed. Data were analyzed by ANOVA and if there is a real difference between the results of the treatment, then the Tukey test. The results showed that the combination of curcumin and vitamin E therapy is able to reduce the expression of IL-1α and TNF-α expression was significantly (p <0.05) with a second therapeutic dose (curcumin 72mg / kg + vitamin E 200 IU) most effective in decreasing the expression of IL-1α (5.76 ± 0.43%) and decreased the expression of TNF-α (6.21 ± 0.67%). The conclusion of this study is the combination of curcumin and vitamin E can be used as a breast cancer therapy
