English Abstract
The hyperactivity of the renin-angiotensin systems (RAS) plays an --_.:)ortant role in hypertension. The most important regulatory component the RAS is angiotensin-converting enzyme (ACE), which converts ziotensin-I (ANG I) to angiotensin-II (ANG II). Recently, ACE inhibitors derived from food proteins are the best-blown group of bioactive peptides and their bio-functionality can be a mnising alternative when choosing between natural bioactive food 71ponents and a synthetic drug. Among red algae, the bioactivities of rochaetium sp used in this study have never been reported. Screening of oboactive peptide from Acrochaetium sp as ACE inhibitor was identified as VGGSDLQAL (using a-Chymotrypsin) and LRDGEIILR (using combination of pepsin — thermolysin). It comes from Phycoerythrin which abundant protein in most red algae. The peptide VL-9 shows the ACE activity with IC50 value 433.1±1.08 pM and the peptide LR-9 has IC50 value 94.28±1.84 pM. The inhibition pattern showed VL-9 as non-competitive inhibitor, while the LR-9 was competitive inhibitor. Molecular docking simulation proved that VL-9 was non-competitive inhibition due to the interaction peptide and ACE was not in the catalytic site and LR-9 was competitive inhibitor due to the interaction of the peptide in the active site. Moreover, VL-9 and LR-9 derived from Acrochaetium sp is a natural bioactive peptide which is safer and available for food protein also the ACE inhibitory peptide derived from Acrochaetium sp could be the one alternative resource to develop functional food for combating hypertension.
