Abstract

Sorgum (Sorghum bicolor) merupakan tanaman serealia yang dikenal adaptif pada lahan kering yang mengandung sedikit nutrisi. Kekurangan nutrisi pada tanah kering umumnya diatasi dengan pemberian pupuk sintetis yang apabila penggunaannya berlebihan akan menurunkan kualitas tanah. Penggunaan agen hayati seperti bakteri menjadi salah satu alternatif untuk mencegah terjadinya kerusakan tanah. Penelitian ini bertujuan untuk mengetahui jumlah sel dan menguji kemampuan isolat bakteri yang diisolasi dari akar (endofit) dan tanah (rhizosfer) sorgum dalam memproduksi IAA, melarutkan fosfat, dan memfiksasi nitrogen. Isolat potensial diidentifikasi secara molekular berdasarkan sekuen 16S rDNA dan diuji interaksinya. Sampel diperoleh dari lahan sorgum Tapos, Bogor. Bakteri penghasil IAA, pelarut fosfat, dan pemfiksasi nitrogen masing-masing diisolasi menggunakan medium TSA yang mengandung triptofan 5 µg/mL, Pikovskaya, dan Nfb. Hasil isolasi didapatkan 42 isolat bakteri penghasil IAA, 20 isolat bakteri pelarut fosfat, dan 26 isolat bakteri pemfiksasi nitrogen. Isolat penghasil IAA, pelarut fosfat dan pemfiksasi nitrogen masing-masing diseleksi menggunakan membran nitroselulosa yang ditetesi dengan reagen Salkowski, pengukuran indeks kelarutan dengan medium Pikovskaya padat, dan SERA Ammonia Test Kit. Hasil seleksi didapatkan masingmasing empat isolat untuk diuji lanjut. Isolat E244 menghasilkan IAA dengan konsentrasi tertinggi 93,97±4,51 µg/mL. Isolat R233 memiliki kemampuan pelarutan fosfat tertinggi 14,02±2,41 µg/mL. Isolat R3-544 memfiksasi nitrogen paling baik dengan konsentrasi amonia 2,83±0,68 µg/mL. Isolat E244, R233, dan R3-544 secara berurutan teridentifikasi sebagai Enterobacter cloacae RCB680 (99,90%), Pantoea dispersa (96,38%), dan Leclercia adecarboxylata (99,80%).

English Abstract

Sorghum (Sorghum bicolor) is a cereal plant which well-known as adaptive plant in arid soil which contain less nutrition. Small amount of nutrition in arid soil generally solved by addition of synthetic fertilizers. The usage of synthetic fertilizer in much amount will decreased soil quality. Biological agent utilization such as bacteria is one of the alternative solution to prevent soil damage. These research aimed to observe the population and potency of bacteria isolated from sorghum root (endophyte) and soil (rhizosphere) on IAA production, phosphate solubilization, and nitrogen fixation. Isolates with the highest potency were identified based on 16S rDNA. The next step was assay the interaction types of isolates obtained. Samples were taken from sorghum plantation in Tapos, Bogor. IAA producing, phosphate solubilizing, and nitrogen fixing bacteria were isolated using TSA medium contained tryptophan 5 µg/mL, Pikovskaya, and Nfb, respectively. Total isolates obtained from isolation were 42 IAA isolates of producing bacteria, 20 isolates of phosphate solubilizer bacteria, and 26 isolates of nitrogen fixing bacteria. Screening of IAA producing was using nitrocellulose membrane with addition of Salkowsky reagent, phosphate solubilization was detected based on solubilization index, and nitrogen fixation potency was assayed using SERA Ammonia Test Kit. Isolate E244 able to produce IAA with highest concentration of 93.97±4.51 µg/mL. Isolate R233 able to solubilizing phosphate with concentration of 14.02±2.41 µg/mL. Isolate R3-544 able to fixing nitrogen with high ammonia concentration of 2.83±0.68 µg/mL. Isolate E244, R233, and R3-544 were identified as Enterobacter cloacae RCB680 (99,90%), Pantoea dispersa (96,38%), and Leclercia adecarboxylata 60ft2 (99,80%) respectively.
