Abstract

Penelitian ini bertujuan menganalisis pengaruh kombinasi ekstrak daun tapak liman (E. scaber) dan katuk (S. androgynus) terhadap sel hematopoietik mencit BALB/c bunting setelah infeksi E. coli. Penelitian terdiri dari 8 kelompok perlakuan dan 3 ulangan, yaitu K+ (kontrol sakit), K- (Kontrol sehat), P1 (infeksi + tapak liman 100%), P2 (infeksi + 75:25% tapak liman dan katuk), P3 (infeksi + 50:50% tapak liman dan katuk), P4 (infeksi + 25: 75% tapak liman dan katuk), P5 (infeksi + katuk 100%), P6 (tanpa infeksi + 50:50% tapak liman dan katuk). Mencit dikawinkan dalam kondisi estrus. Kombinasi ekstrak diberikan secara oral gavage pada hari ke- 1 hingga ke-4 kebuntingan. Mencit diinfeksi E. coli 107 CFU/ml dalam 0,1 ml secara intraperitonial pada hari ke-5 kebuntingan. Kombinasi ekstrak diberikan hingga hari pembedahan. Pembedahan bone marrow dilakukan pada hari ke-8 dan hari ke-16 kebuntingan. Sel hematopoietik diisolasi dengan cara flushing out. Sel diwarnai dengan antibodi spesifik dan dianalisis dengan flowcytometri. Data lalu dianalisis dengan two-way ANOVA dengan taraf signifikan 95%. dengan uji lanjut tukey test. Hasil menunjukkan kombinasi ekstrak belum mampu menigkatkan ekspresi sel GR1+. Kelompok P1 (tapak liman 100%) berpengaruh di hari ke-16 kebuntingan pada sel TER119+VLA4+. Kelompok P5 (katuk 100%) meningkatkan sel B220+SDF-1+ pada hari ke-8 kebuntingan serta sel TER119+CD34+ pada hari ke-16 kebuntingan.

English Abstract

The aim of this research is to analyze the influence of the combination of tapak liman extract (E. scaber) katuk extract (S. androgynus) to hematopoietic cells of BALB/c pregnant mice after E. coli infection.. The study consisted of 8 treatment groups with 3 replications, K+ (Sick control), K- (healthy control), P1 (infection + tapak liman 100%), P2 (infection + 75:25% tapak liman and katuk), P3 (infection + 50:50% tapak liman and katuk), P4 (infection + 25: 75% tapak liman and katuk), P5 (infection + katuk 100%), P6 (without infection + 50:50% tapak liman and katuk). Mice mated in estrus condition. The combination of extracts was administered on oral gavage on day 1 to 4 days of pregnancy. Mice were then infected with E. coli of 107 CFU / ml in 0.1 ml by intraperitoneally on the 5th day of pregnancy. The extract combination was given back until dissection. Bone marrow dissection is performed on, 8th, and 16th days of pregnancy. The hematopoietic cells were isolated by flushing out the bone marrow. Cells were then stained with specific antibodies and analyzed with flowcytometry. The data were then analyzed by two-way ANOVA with a significant level of 95% and then tested by tukey test. The results show that the combination of extracts has not been able to increase the expression of GR1+ cells. The P1 group (100% of tapak liman) had an effect on the 16th day of pregnancy on TER119+VLA4+ cells. The P5 group (100% of katuk) increased B220+SDF-1+ cells on the 8th day of pregnancy and TER119+CD34 + cells on the 16th day of pregnancy.
