Abstract

Cabai G1M1 merupakan salah satu tanaman cabai rawit mutan hasil induksi mutasi dengan ethyl methane sulfonate (EMS) 0,01% terhadap cabai rawit Genotipe 1 (G1). Tujuan penelitian ini adalah mengevaluasi variasi karakter morfologi, menganalisis kandungan capsaicin, dan profil genetik gen KasI antara tanaman cabai rawit mutan G1M1 dan G1 kontrol. Pengamatan morfologi dilakukan ketika tanaman berumur 120 HST. Karakter yang diamati meliputi tinggi tanaman, jumlah cabang, hari pembungaan pertama, jumlah bunga pada setiap aksil, panjang dan diameter buah, jumlah biji, dan berat buah. Kandungan capsaicin diukur dari buah berumur antara 30-35 HSP, metode yang digunakan adalah metode spektrofometri (Ȧ280 nm). Profil gen KasI pada tanaman mutan dan kontrol dianalisis dengan metode sekuensing dan hasilnya akan disejajarkan serta dibandingkan dengan sekuens Capsicum annuum 3-oxoacyl- (acyl-carrier-protein) synthase (KasI) gene (KM037709.1) pada NCBI. Hasil penelitian ini menunjukkan bahwa tanaman mutan G1M1 memiliki karakter generatif dan tinggi tanaman yang berbeda signifikan dengan tanaman G1 kontrol, sedangkan karakter jumlah cabang, jumlah bunga pada setiap aksil, dan hari pembungaan mutan G1M1 tidak berbeda nyata dengan G1 kontrol. Kandungan capsaicin antara tanaman G1M1 tidak berbeda signifikan dengan kontrol, akan tetapi kandungan capsaicin G1M1 cenderung lebih tinggi dibandingkan dengan kontrol. Gen KasI mutan G1M1 mengalami mutasi titik berupa subtitusi. Subtitusi ini menyebabkan terjadinya perubahan komposisi asam amino. Hal ini kemungkinan mengakibatkan peningkatan variasi kandungan capsaicin pada tanaman mutan G1M1

English Abstract

Cayenne pepper G1M1 is one of mutant plant produced from cayenne pepper Genotype 1 (G1) induced mutation with 0.01% ethyl methane sulfonate (EMS). The aim of this research is to evaluate the variation of morphological characters, analyze of capsaicin content, and genetic profile of KasI gene between G1M1 mutant and G1 control. Morphological observations were performed at 120 DAP (day after planting). The observed characters included plant height, number of branches, first flowering, number of flowers on each axillar, length and diameter of the fruit, number of seeds, and weight of fruit. Capsaicin content measured from fruit at 30-35 DPA (day post anthesis) using spectrophotometry method (Ȧ280 nm). Genetic profile of KasI genes of mutant plants and controls was analyzed by sequencing method and the results aligned and compared with the sequnce of Capsicum annuum KasI (KM037709.1) on the NCBI. The results of this study indicate that G1M1 mutant plant has generative characters and plant height that significantly different with G1 control, while the character number of branch, the number of flowers on each axis, and the day of flowering G1M1 mutant is not significantly different with G1 control. Capsaicin content between G1M1 plants did not differ significantly with G1 control, but the capsaicin content of G1M1 tended to be higher than control. The KasI gene of G1M1 mutant shows the presence of substitution at some sites. This extinction causes the replacement of amino acid composition, this is one of possibility the causes of differences in the capsaicin content between plants.
