Abstract

Kondisi hamil mengakibatkan terjadinya berbagai perubahan fisiologis dan hormonal sehingga lebih rentan terinfeksi bakteri yang dapat diminimalkan dengan konsumsi herbal. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian kombinasi ekstrak etanol daun katuk (S. androgynus) dan tapak liman (E. scaber) terhadap histologi ginjal dan hepar mencit bunting terinfeksi E. coli. Hewan coba dibagi delapan perlakuan yaitu kontrol sehat (K-), kontrol sakit (K+), P1, P2, P3, P4, P5, dan P6 (perlakuan kombinasi ekstrak tanpa infeksi). Mencit bunting diberi ekstrak secara oral pada kebuntingan hari ke-1 sampai hari ke-4. Hari ke-5 kebuntingan mencit diinfeksi E. coli 107 CFU/ml secara intraperitoneal. Setelah infeksi dilanjutkan pemberian ekstrak hingga hari pembedahan yaitu hari ke-8 dan ke-16 kebuntingan. Organ ginjal dan hati diisolasi dan dibuat preparat parafin dengan pewarnaan HE (Hematoxylin-Eosin). Selanjutnya sel nekrosis dihitung dan dianalisa menggunakan two way ANOVA dengan selang kepercayaan 95 % kemudian dilanjutkan Tukey test. Hasil nilai nekrosis masing-masing perlakuan kombinasi ekstrak dibandingkan dengan K+ mengalami penurunan yang berbeda nyata. Sedangkan nilai nekrosis P6 dibandingkan dengan K- mengalami peningkatan yang berbeda nyata. Hal ini menunjukkan pemberian kombinasi ekstrak pada keadaan normal dapat meningkatkan nekrosis namun dalam keadaan terinfeksi dapat menurunkan nekrosis. Bunting hari ke-8 nekrosis ginjal terendah P1 (23,00%) sedangkan nekrosis hepar terendah P5 (20,60%). Bunting hari ke-16 nekrosis ginjal terendah P2 (28,73%) sedangkan nekrosis hepar terendah P5 (33,40%). Berdasarkan hasil tersebut dosis P1 dan P5 merupakan dosis yang paling aman untuk dikonsumsi.

English Abstract

Pregnant conditions result in various physiological and hormonal changes, so it make more susceptible to bacterial infections that can be minimized by herbal consumption. The aim of this research is to know the effect of combination ethanol extract tapak liman leaf (E. scaber) and katuk leaf (S. androgynus) for kidney and hepar histology of pregnant mice that infected E. coli. The animals trials were divided into eight groups: healty control (K-), diseased control (K+), P1, P2, P3, P4, P5, and P6 (treatment combination extract without infected E. coli). Pregnant mice were given extract by oral on pregnancy day 1th to day 4th. The 5th day of pregnancy mice were intraperitoneally infected with E. coli. Then continued giving extract until surgery in day 8th, and day 16th pregnancy. Kidney and hepar organ was isolated and then the isolated organs are prepared with paraffin and HE (Hematoxylin-Eosin) staining. The calculation of necrosis cells in each preparat analyzed using two way ANOVA with convidence interval 95% and continued with Tukey test. The results of necrosis values in each treatment combination extracts compared with K+ decrease with different significantly. While the necrosis values of P6 compared with K- increase with different significantly. This suggests that combination extracts in normal may increase necrosis but in an infected state may decrease necrosis. In 8th day pregnancy lowest kidney necrosis is P1 (23,00%) and lowest hepar necrosis is P5 (20,60%). In 8th day pregnancy lowest kidney necrosis is P2 (28,73%) and lowest hepar necrosis is P5 (33,40%). Based on these results, the doses of P1 and P5 are the safest doses to be consumed.
