Abstract

Penelitian ini bertujuan untuk mengetahui pengaruh kompleks scavenger AOC (Aspirin-Ovalbumin-Caffeine) terhadap distribusi MDA pada jaringan mukosa lambung tikus model gastritis induksi aspirin. Dua puluh tiga tikus betina Wistar dibagi menjadi 8 kelompok perlakuan, yaitu (1) kelompok kontrol negatif (K-); (2) kelompok AOC; (3) gastritis akut pada tikus umur 1,5 bulan; (4) gastritis akut pada tikus umur 6 bulan; (5) gastritis diberi AOC umur 1,5 bulan; (6) gastritis diberi AOC umur 6 bulan; (7) gastritis tanpa AOC umur 1,5 bulan; (8) gastritis tanpa AOC umur 6 bulan. Gastritis akut diinduksi dengan pemberian aspirin (300 mg/kg BB) secara per oral. Kerusakan mukosa lambung dinilai berdasarkan metode Coleman. Distribusi malondialdehid (MDA) dideteksi menggunakan imunohistokimia (IHK). Erosi dan spot perdarahan pada mukosa lambung tikus mengindikasikan adanya kerusakan pada mukosa lambung. Erosi dan spot perdarahan paling banyak, dengan skor sebesar 5,3, ditunjukkan oleh kelompok gastritis akut umur 1,5 bulan. Penurunan skor erosi dan spot perdarahan terjadi pada tikus gastritis yang diberi AOC. Tikus yang diberi AOC saja secara menarik tidak menunjukkan tanda-tanda kerusakan mukosa lambung berupa erosi atau spot perdarahan. Distribusi MDA dalam jaringan mukosa lambung meningkat secara signifikan pada kelompok gastritis akut umur 1,5 dan 6 bulan (masingmasing 83,64% ± 4,37 dan 49,71% ± 5,36) dibandingkan dengan kelompok gastritis (+) AOC umur 1,5 dan 6 bulan (masing-masing 4,39% ± 2,95 dan 0,62% ± 0,40) dan AOC (0,64% ± 0,03). Hasil penelitian menunjukkan bahwa kompleks scavenger AOC dapat menurunkan jumlah erosi dan spot perdarahan serta distribusi MDA, sehingga kompleks AOC dapat digunakan sebagai anti-gastritis.

English Abstract

This research aims to know the effect of complex AOC scavenger (Aspirin-Ovalbumin-Caffeine) on the distribution of MDA in gastric mucosal tissue of aspirin-induced gastritis model. Twenty-three Wistar female rats were divided into 8 treatment groups, that is (1) negative control group (K-); (2) AOC group; (3) acute gastritis in 1.5 month-old rat; (4) acute gastritis in 6 month-old rat; (5) gastritis treated with AOC in 1.5 month-old rat; (6) gastritis treated with AOC in 6 month-old rat; (7) gastritis without AOC in 1.5 month-old rat; (8) gastritis without AOC in 6 month-old rat. Acute gastritis is induced by oral administration of aspirin (300 mg/kg BW). Gastric mucosal damage is assessed by the Coleman method. Malondialdehyde distribution (MDA) is detected using immunohistochemistry (IHC). Erosion and spot bleeding in gastric mucosal of rat indicate a damage in the gastric mucosa. The highest number of erosion and bleeding spot, with a score of 5.3, was shown by the acute gastritis group of 1.5 month-old rat. Decreased scores of erosion and bleeding spot was shown in AOC-treated gastritis rat. The rat treated with AOC alone did not show any signs of gastric mucosal damage in the form of erosion or bleeding spot. MDA distribution in gastric mucosal tissue increased significantly in the acute gastritis group of 1.5 and 6 months (respectively 83.64% ± 4.37 and 49.71% ± 5.36) compared with the AOC-treated gastritis group age of 1.5 and 6 months (respectively 4.39% ± 2.95 and 0.62% ± 0.40) and AOC (0.64% ± 0.03). The results showed that complex of AOC scavenger can decrease the amount of erosion and bleeding spot as well as MDA distribution, so that AOC complex can be used as anti-gastritis.
