Abstract

Vaksinasi campak merupakan salah satu upaya pencegahan penyakit campak dengan memberikan antigen virus campak yang telah dilemahkan. Tujuan dari vaksinasi untuk menstimulasi aktivasi respon imun seluler dan humoral. Adanya gangguan respon imun akibat polutan Benzo[a]pyrene (BaP) yang memiliki efek imunosupresi dapat menyebabkan kerja sistem imunitas tubuh dalam merespon infeksi penyakit juga menjadi terganggu. Penelitian ini dilakukan untuk mengetahui pengaruh BaP terhadap profil sel T helper CD4+, T CD4+CD25+ dan TGF-β pada organ spleen mencit pasca injeksi vaksin campak secara in vivo berdasarkan perhitungan jumlah relatif sel melalui analisis flowcytometry. Marker antibodi spesifik yang digunakan antara lain anti-CD4, anti-CD25 and anti-TGF-β. Hasil penelitian menunjukkan bahwa BaP (20 mg/kg) mampu menurunkan jumlah relatif sel T CD4+ helper, namun efek supresi BaP tidak bekerja pada sel T CD4+CD25+ dan TGF-β. Hal tersebut ditandai dengan adanya peningkatan jumlah T CD4+CD25+ dan TGF-β secara signifikan setelah diberi BaP pasca vaksinasi campak. Penurunan sel T CD4+ helper serta kenaikan T CD4+CD25+ dan TGF-β kemungkinan terjadi karena keterlibatan AhR. Sel T helper CD4+ berperan penting dalam aktivasi sistem imunitas untuk produksi antibodi dan diferensiasi sel-sel efektor, sedangkan T CD4+CD25+ dan TGF-β berperan dalam regulasi sistem imunitas khususnya respon inflamasi melalui mekanisme supresi. Adanya penurunan sel T helper CD4+ (produksi antibodi dan peran sel efektor T menurun) atau peningkatan jumlah T CD4+CD25+ dan TGF-β (supresi sel imun lain meningkat) akibat pemberian BaP secara terusmenerus, kemungkinan dapat berpengaruh terhadap pemberian vaksin campak dan mengganggu keberhasilan vaksinasi.

English Abstract

The measles vaccination is a prevention of measles disease by giving the weakened measles virus into the body. The purpose of vaccination is stimulating the activation of cellular and humoral immune responses. Benzo[a]pyrene (BaP) is a pollutant that has immunosuppressive effect. This pollutant can affect activity of immune systems to eliminate and clear foreign antigen in the body. This research has an object to determine the effect of BaP on the profil of CD4+ helper T cell, CD4+CD25+ T cell and level of TGF-β in spleen after exposure of BaP (20 mg/kg) pasca measles vaccination (20 mg/kg), using flow cytometry stained with spesific marker anti-CD4, anti-CD25 and anti-TGF-β. The results showed that BaP (20 mg/ kg) was able to decrease the relative amount of CD4+ helper T cells, but the suppression effect of BaP did not work on CD4+CD25+ T cells and TGF-β. Data shows that CD4+CD25+ T cells and TGF-β significantly decreased after exposure of BaP pasca measles vaccination. Supression of CD4+ helper T-cell and increase of CD4+CD25+ T cells and TGF-β probably involve the interaction between BaP ligand with its receptor, AhR. CD4+ helper T cells have important function in immune system, especially for antibody production and differentiation of effector cells in vaccination, whereas CD4+CD25+ T cells and TGF-β have regulation function to other immune cells. Reduction of CD4+ helper T cells can lead to decreased production of antibodies and T cell response. Rise of CD4+CD25+ T cells and TGF-β will increase immune suppression. Because of those two reasons, long exposure of BaP probably will affect effectiviteness of measles vaccine.
