Abstract

Tanaman G1M8 merupakan generasi kedua tanaman cabai rawit Genotipe 1 (G1) yang telah diinduksi Ethyl Methanesulfonate (EMS) 0,01%. Tanaman tersebut perlu dinalisis berdasarkan morfologi, fisiologi dan molekuler. Tujuan penelitian ini yaitu mengetahui variasi morfologi, kandungan capsaicin, serta polimorfisme gen pAMT antara tanaman mutan G1M8 dan G1 kontrol (tidak diinduksi EMS). Analisis morfologi yang meliputi tinggi tanaman, jumlah cabang, waktu pembungaan, panjang, diameter, dan berat buah, jumlah biji, serta warna antera, mahkota bunga, dan buah muda dilakukan saat tanaman berumur 92 HST. Kandungan capsaicin diukur dari filtrat sampel buah cabai G1M8 dan G1 kontrol berumur 30-40 HSP menggunakan metode spektrofotometri pada panjang gelombang 280 nm. Analisis data morfologi dan kandungan capsaicin dilakukan dengan uji T tidak berpasangan dan Mann-Whitney. Analisis molekuler dilakukan dengan sekuensing gen pAMT, kemudian dianalisis menggunakan metode ClustalW Multiple Alignment. Hasil alignment dibandingkan dengan sekuen gen pAMT Capsicum annuum cv. Himo (kode genebank: LC032105.1). Sebagian besar tanaman G1M8 memiliki karakter morfologi dan kandungan capsaicin yang lebih unggul dibandingkan G1 kontrol. Variasi genetik pada gen pAMT tanaman G1M8 dan G1 kontrol ditunjukkan oleh adanya perubahan basa nukleotida pada ekson dan intron. Mutasi pada ekson 3 gen pAMT tanaman G1M8/3 membentuk stop kodon, sehingga kandungan capsaicinnya lebih rendah daripada G1 kontrol. Variasi genetik tanaman dapat ditunjukkan oleh variasi morfologi, fisiologi dan sekuen gen tanaman.

English Abstract

The G1M8 plants is a second generation of Genotipe 1 (G1) cayenne pepper plant induced with 0,01% Ethyl Methanesulfonate (EMS). The plants need to be analyzed based on its morphological, physiological and molecular characters. The purpose of this research is to analyze the difference of morphological characters, capsaicin content, and polymorphism of pAMT gene between G1M8 and G1 control (not induced with EMS). The morphological characters including plant height, number of branches, day of flowering, length, diameter, and weight of fruit, number of seeds, anther color, flower crown color, and immature fruit color was analyzed at 92 DAP. The capsaicin content was measured from filtrate samples of 30-40 DPA fruit of G1M8 and G1 control using spectrophotometry method at 280 nm wavelength. Morphological characters and capsaicin content data were analyzed with T-indepedent and Mann-Whitney test. Molecular analysis was performed by sequencing of pAMT gene, then analyzed using ClustalW Multiple Alignment methods and compared with the sequence of pAMT gene of Capsicum annuum cv. Himo (genebank code: LC032105.1). Most of G1M8 plants have higher value of morphological characters and capsaicin content than G1 control. Genetic polymorphism occurred by nucleotide base alterations in exons and introns of pAMT gene of G1M8 dan G1 control. Mutation in pAMT gene exon 3 of G1M8/3 formed stop codon, so its capsaicin content was lower than G1 control’s. Genetic variation of plants are showed by their variations of morphology, physiology and plant gene sequences.
