Abstract
Studi terbaru menyatakan kanker payudara dapat mempengaruhi fisiologi organ lain, termasuk stres oksidatif pada hati. Beberapa protein seperti HO, SOD dan Nrf2 diketahui mempunyai peran penting dalam menetralisir oksidan dalam hati. Selain itu sitokin proinflamasi seperti TNF-α dan IFN-γ juga banyak berperan dalam patogenesis penyakit hati. Pada sisi lain beberapa bahan seperti Temu Putih (Curcuma zedoaria) dan Meniran (Phyllantus urinaria) mempunyai kemampuan antioksidan dan imunomodulator baik in vitro maupun in vivo. Kedua tumbuhan tersebut kemudian digabungkan dalam bentuk ramuan Cheral. Tujuan penelitian ini adalah mengetahui pengaruh pemberian Cheral dalam memodulasi stres oksidatif pada hati mencit model kanker payudara. Induksi kanker payudara dilakukan dengan injeksi DMBA pada daerah sekitar areola mencit. Level protein Nrf2, HO, SOD dari hepatosit dan sitokin proinflamasi TNF-α dan IFN-γ dari limfa dianalisis dengan flow sitometri setelah pemberian Cheral selama 14 hari. Analisis data dilakukan dengan ANOVA satu arah dan uji Tukey HSD (α=5%). Mencit model kanker payudara mengalami stres oksidatif berdasarkan tingginya level Nrf2, HO dan SOD pada kelompok kanker. Tingginya level sitokin TNF-α dan IFN- γ pada kelompok kanker menunjukkan adanya kontribusi kedua sitokin tersebut dalam menghasilkan kondisi stres oksidatif. Pemberian ramuan Cheral mampu memodulasi kondisi redoks dengan menurunkan level HO dan SOD, namun tidak pada Nrf2, begitu juga dengan level TNF-α dan IFN-γ. Uji korelasi menunjukkan adanya pengaruh level TNF-α dan IFN- γ dari limfa terhadap level HO dan SOD pada hati. Penurunan level TNF- α dan IFN-γ juga turut berkontribusi untuk menyeimbangkan kondisi reduksi-oksidasi dalam hati. Hasil ini menunjukkan bahwa ramuan Cheral berpotensi sebagai terapi alternatif untuk mengatasi stres oksidatif pada hati akibat kanker payudara.
English Abstract
Recent studies report that breast cancer may affect the physiology of other organs, including oxidative stress in the liver. Some proteins such as HO, SOD, and Nrf2 are known to play an important role in balancing redox conditions in the liver. In addition, proinflammatory cytokines such as TNF-α and IFN-γ also play a significant role in the pathogenesis of liver disease. On the other hand, some ingredients such as Temu Putih (Curcuma zedoaria) and Meniran (Phyllantus urinaria) have ability to maintain redox stability and immunomodulation both in vitro and in vivo. Both of them are combined in the form of Cheral potion. The aim of this research is to analyze the effect of Cheral in modulating oxidative stress in the liver of breast cancer model mice. In vivo experiment was performed using DMBA-induced mammary cancer mice. Nrf2, HO and SOD were measured from hepatocyte, meanwhile TNF-α and IFN-γ were measured from splenocyte. Nrf2, HO, SOD, TNF-α, and IFN-γ level were analyzed by flow sitometry after 14 days of Cheral administration. Data analysis with one way ANOVA and Tukey HSD to determine significant difference among groups (α=5%). The results showed that mice model of breast cancer underwent oxidative stress indicated by high levels of Nrf2, HO, and SOD. High levels of TNF-α and IFN-γ in the cancer group indicated the contribution of both cytokines in disturbing redox homeostasis. In contrast, Cheral treatment was able to modulate redox balance by decreasing HO and SOD levels, but not for Nrf2, as well as TNF-α and IFN-γ levels. Pearson correlation test indicated there were correlation between TNF-α and IFN-γ level from spleen with HO and SOD level from liver. Therefore, decreasing of TNF-α and IFN-γ levels also contribute to normalize redox condition in the liver. These results showed that Cheral has potency as an alternative therapeutic agent to reduce oxidative stress in the liver under breast cancer condition.
