Abstract
Klebsiella pneumoniae merupakan salah satu bakteri gram negatif yang sering menyebabkan infeksi pada manusia. Tidak hanya menyebabkan penyakit pneumonia, Klebsiella pneumoniae juga sering menyebabkan infeksi nosokomial. Oleh karena itu perlu dilakukan penelitian untuk mendapatkan antimikroba seperti dari bahan-bahan alami. Salah satunya adalah esktrak kulit salak pondoh (Salacca zalacca) yang memiliki kandungan flavonoid, tannin dan saponin. Penelitian ini bertujuan untuk mengetahui pengaruh ekstrak kulit salak pondoh (Salacca zalacca) terhadap pertumbuhan bakteri Klebsiella pneumoniae. Metode yang digunakan dalam penelitian ini adalah dilusi agar untuk menentukan Kadar Hambat Minimum (KHM). Ekstrak kulit salak pondoh dibuat dengan esktraksi maserasi dengan menggunakan etanol 96%. Konsentrasi ekstrak yang digunakan yaitu 0%, 2,5%, 5%, 7,5%, 10% dan 12,5%. Setiap perlakuan dilakukan pengulangan sebanyak 4 kali. Kadar hambat bakteri ditentukan dengan menghitung langsung tanpa alat. Hasil uji statistik Kruskal Wallis menunjukkan adanya perbedaan signifikan antara ekstrak etanol kulit salak pondoh dengan bakteri Klebsiella pneumoniae karena setiap ekstrak memiliki p(0,000) < 0,05. Pada uji Man Whitney menunjukkan adanya perbedaan yang signifikan pada setiap perbandingan konsentrasi ekstrak kulit salak pondoh karena memiliki p value < 0,05. Pada uji Kolerasi Spearman, hubungan antara konsentrasi kulit salak pondoh terhadap pertumbuhan koloni bakteri termasuk ke dalam hubungan yang sangat kuat. Berdasarkan hasil penelitian dapat disimpulkan bahwa ekstrak kulit salak pondoh memiliki efek antimikroba terhadap Klebsiella pneumoniae dengan Kadar Hambat Minimum (KHM) 12,5%
English Abstract
Klebsiella pneumoniae is one of the gram-negative bacteria that most commonly causes infections in humans. Not only causes pneumonia, Klebsiella pneumoniae also often causes nosocomial infections. Therefore, it is necessary to conduct research to obtain alternative antimicrobials such as natural ingredients. One of them is Salak Pondoh peel extract (Salacca zalacca) which contains flavonoids, tannins and saponins. This study aims to determine the effect of Salak Pondoh peel (Salacca zalacca) on the growth of Klebsiella pneumoniae bacteria. The method used in this study is agar dilution to determine the Minimum Inhibitory Level (MIC). Salak Pondoh peel extract was made with maceration extraction using 96% ethanol. The concentrations of extracts were 0%, 2.5%, 5%, 7.5%, 10% and 12.5%. Each treatment was repeated 4 times. Bacterial inhibitory levels by counting directly without tools. The results of the Kruskal Wallis statistic test showed that there were significant ethanol extracts of salak pondoh skin with Klebsiella pneumoniae bacteria because each extract had p (0,000) <0.05. In the Man Whitney test showed that there was a significant difference in each alternative concentration of salak pondoh skin extract because it had a p value <0.05. In the Spearman coleration experiment, the relationship between skin salak pondoh concentration on the growth of bacterial colonies was included in a very strong relationship. Based on the results of the study it can be concluded that salak pondoh skin extract has antimicrobial effects on Klebsiella pneumoniae with a minimum inhibitory level (MIC) of 12.5%.
