Abstract
Latar Belakang: Ulkus gaster merupakan keadaan terjadinya kerusakan mukosa lambung sampai lapisan otot akibat inflamasi. Teh hijau yang telah diketahui memiliki banyak manfaat dalam dunia kesehatan mengandung epigallocatechin- 3-gallate (EGCG) yang dapat dipergunakan sebagai antiinflamasi dengan mekanisme penghambatan COX-2 dan mediator inflamasi seperti TNF-α, IL-1β dan IFN-γ. Tujuan penelitian ini untuk mengetahui efek ekstrak air teh hijau terhadap gambaran histopatologi epitel mukosa lambung tikus yang diinduksi indometasin. Metode: Penelitian ini merupakan studi eksperimental dengan rancangan post-test only, control group design menggunakan 25 hewan coba tikus strain Wistar jantan yang dibagi menjadi 5 kelompok, yaitu kelompok kontrol negatif, kelompok kontrol positif yang diinduksi indometasin 30 mg/kgBB dan 3 kelompok perlakuan yang diinduksi indometasin 30 mg/kgBB dan pada kelompok perlakuan diberi ekstrak air teh hijau 50 mg/kgBB, 100 mg/kgBB dan 200 mg/kgBB berturut – turut. Penilaian gambaran kedalaman lesi histopatologi epitel mukosa lambung tikus menggunakan modifikasi sistem skoring oleh Sibilia et al. Hasil: menunjukan adanya perbedaan signifikan pada gambaran histopatologi epitel mukosa lambung setelah diberi ekstrak air teh hijau (Kruskal Wallis, p=0.000). Hasil uji Mann Whitney menunjukan perbedaan yang signifikan antara kelompok kontrol negatif dengan keempat kelompok lainya serta antara kelompok kontrol positif dengan kelompok perlakuan dengan dosis ekstrak air teh hijau 50 mg/kgBB (p<0,05). Hasil yang tidak signifikan didapatkan antara kontrol positif dengan kelompok yang diberi ekstrak air teh hijau 100mg/kgBB dan 200 mg/kgBB. Hasil uji spearman didapatkan koefisien korelasi 0,125 (p>0,05). Kesimpulan: ekstrak air teh hijau dapat memperbaiki gambaran histopatologi epitel mukosa lambung tikus yang diinduksi indometasin pada dosis 50 mg/kgBB
English Abstract
Background: Gastric ulcer is defects in the gastric mucosa that extend through the muscularis mucosa which is caused by inflammation. Green tea has been well-known for having many benefits in the medical field. It contains epigallocatechin-3-gallate (EGCG), which is known to have anti-inflammatory effect by inhibition of COX-2 and inflammation mediators such as TNF-α, IL-1β and IFN-γ. The purpose of this experiment is to investigate the effect of aqueous green tea leaf extract on gastric mucosa histopathology of rats induced by indomethacin. Methods: This experiment was conducted with post test only control group design on 25 wistar strain male rats and divided into 5 group, which were negative control group, positive control group and 3 treatment groups. The three treatment groups were induced by indomethacin 30mg/kgBW, and given aqueous green tea extract, at 50mg/kgBW, 100mg/kgBW, and 200mg/kgBW respectively. The lesion depth of gastric mucosa was evaluated with modification of scoring system developed by Sibilia et al. Results: a significant difference in gastric mucosa epithelial histopathology after being given green tea aqueous extract (Kruskal Wallis, p=0.000). Mann Whitney Test showed a significant difference between the negative control group and the other 4 groups, also between the positive control group and the treatment group at the dose of 50 mg/kgBW aqueous green tea extract (p< 0.05). There was no significant difference between the positive control group and the other 2 treatment groups (100 mg/kgBW and 200 mg/kgBW). Spearman correlation test shows the correlation coefficient is 0.125 (p>0.05). Conclusions: aqueous green tea extract can repair gastric mucosa epithelial histopathology of rats induced by indomethacin at a dose of 50mg/kgBW.
