Abstract
Cocor bebek (Bryophyllum pinnatum) lazim dipakai di masyarakat sebagai ramuan tradisional penurun panas, pengobatan wasir, antibatuk dan antiinflamasi. Pembuktian secara ilmiah mengenai keamanannya penting untuk diketahui, salah satunya melalui uji toksistas akut. Tujuan dari penelitian ini adalah untuk mengetahui efek toksisitas akut esktrak cocor bebek pada tikus strain Wistar yang diukur secara kuantitatif dengan LD50. Penelitian ini merupakan studi eksperimental dengan rancangan post-test only, control group design menggunakan 30 hewan coba tikus strain Wistar jantan yang dibagi menjadi 6 kelompok, yaitu kelompok kontrol yang hanya diberi akuades dan 5 kelompok perlakuan yang diberi ekstrak etanol daun cocor bebek 450 mg/kgBB, 900 mg/kgBB, 1800 mg/kgBB, dan 3600 mg/kgBB berturut-turut dengan 5000 mg/kgBB pada 6 tikus pada kelompok kontrol di pertengahan penelitian. Ekstrak etanol daun cocor bebek diberikan secara oral dengan hanya satu kali pemberian pada awal masa penelitian. Nilai LD50 ekstrak etanol daun cocor bebek adalah lebih besar dari 5000 mg/kgBB dengan tidak ditemukannya gejala toksisitas akut pada hewan coba selama pengamatan sesuia kriteria Loomis (1978). Sebagai kesimpulan, ekstrak etanol daun cocor bebek adalah bahan yang praktis tidak toksik berdasarkan kriteria Loomis (1978) dan tidak ada gejala klinis toksisitas akut yang signifikan yang terjadi pada seluruh hewan coba. Berdasarkan penelitian yang sudah dilakukan, perlu dilakukan penelitian lebih lanjut untuk meneliti potensi toksisitas subkronis dan kronis dari ekstrak cocor bebek dengan jumlah hewan coba yang lebih banyak dan rentang dosis yang lebih bervariasi.
English Abstract
Cocor bebek (Bryophyllum pinnatum) is commonly used in the community as a traditional febrifuge, treatment of hemorrhoids, anti-coughing and anti-inflammatory. Scientific proof of its safety is important to know, one of which is through the acute toxicity test. The purpose of this study was to determine the effect of acute toxicity of duck coco extracts on Wistar strain rats measured quantitatively with LD50. This study was an experimental study with a post-test only design, control group design using 30 experimental animals of male Wistar strain rats which were divided into 5 groups, namely the control group which was only given aquadest and 4 treatment groups which were given ethanol extract of cocor bebek leaves 450 mg/kgBW, 900 mg/kgBW, 1800 mg/kgBW, and 3600 mg/kgBW, respectively, then 5000 mg/kgBW to the first 6 rats (control group) in the middle of experiment. Ethanol extract of cocor bebek leaves is given orally with only one administration at the beginning of the study period. LD50 value of ethanol extract of cocor bebek leaves is greater than 5000 mg/kgBW with no symptoms of acute toxicity based on Loomis criteria (1978) in experimental animals during observation. In conclusion, the ethanol extract of cocor bebek leaves is practically non-toxic based on Loomis (1978) criteria and no significant clinical symptoms of acute toxicity that occur in all experimental animals. Based on the research that has been done, further research is needed to investigate the potential for subchronic and chronic toxicity of cocor duck extract with more number of experimental animals and a more varied dose range.
