Abstract

Perkembangan industrialisasi global berhubungan dengan peningkatan angka obesitas, yang secara tidak langsung juga menyebabkan peningkatan prevalensi sindrom metabolik secara dramatis. Manifestasi sindrom metabolik di hepar adalah penyakit hati berlemak non alkoholik. Hati yang berlemak menyebabkan peningkatan peroksidasi lemak dan produknya yaitu malondialdehida (MDA) yang memicu terjadinya reaksi inflamasi lebih lanjut. Dunaliella sp. merupakan mikroalga hijau yang banyak ditemukan di laut Indonesia dan mengandung karotenoid yang tinggi. Tujuan penelitian ini adalah untuk mengetahui pengaruh pemberian ekstrak Dunaliella sp. terhadap kadar MDA hepar tikus model sindrom metabolik. Tikus wistar jantan yang diinduksi diet aterogenik dan minum fruktosa 20% menunjukkan peningkatan kadar MDA hepar secara signifikan (p<0,05) bila dibandingkan kontrol negatif. Pemberian ekstrak metanol Dunaliella sp. dosis 500 mg/kgBB, 1000 mg/kgBB, dan 1500 mg/kgBB secara signifikan menurunkan kadar MDA hepar (p<0,05). Hasil penelitian ini membuktikan bahwa dosis ekstrak berkorelasi negatif dengan kadar MDA hepar dan tingkat keeratan hubungannya sangat kuat (r = -0,836). Kesimpulan yang diperoleh adalah ekstrak metanol Dunaliella sp. dapat menurunkan kadar MDA hepar tikus model sindrom metabolik secara signifikan.

English Abstract

The development of global industrialization is associated with increasing rates of obesity, which also indirectly causes an increase in the prevalence of metabolic syndrome dramatically. The manifestation of metabolic syndrome in the liver is non-alcoholic fatty liver disease. A fatty liver causes an increase in lipid peroxidation and its product, malondialdehyde (MDA) which triggers further inflammatory reactions. Dunaliella sp. is a green microalgae that widely found in the Indonesian sea and it contains high carotenoids. The purpose of this study was to determine the effect of Dunaliella sp. methanol extract against MDA levels in liver models of metabolic syndrome. Male wistar rats which was induced with atherogenic diet and 20% fructose show significantly increased liver MDA levels (p <0.05) when compared to negative controls. Treatment of methanol extract Dunaliella sp. with doses of 500 mg/kg body weight, 1000 mg/kg body weight and 1500 mg/kg body weight significantly decrease liver MDA levels (p <0.05). The results of this study prove that the extract dose is negatively correlated with MDA levels of the liver and the degree of closeness is very strong (r = -0,836). The conclusions is Dunaliella sp. methanol extract. can significantly reduce liver MDA levels in rats with metabolic syndrome.
