Abstract

Diabetescmelitus merupakancpenyakit kronis yang disebabkan defisiensi produksicinsulin atau ketidakmampuan tubuh menggunakan insulin secara efektif. Diabetesimelitus yangctidak terkontrol dapat menyebabkan komplikasi yaitu ulkus diabetes. Komplikasi ini diakibatkan karena adanya gangguan pembuluh darah perifer karena peningkatan Reactive Oxygen Species (ROS). Peningkatan ROS menyebabkan penurunan faktor pertumbuhan seperti TGF-β, sehingga menyebabkan penurunan sintesis kolagen, epitelisasi juga angiogenesis dan menghambat penutupan luka. Tujuan penelitian ini untuk mengetahui efek ekstrak ubi jalar ungu topikal terhadap persentase penutupan luka tikus Rattus norvegicus strain wistar model diabetes. Peningkatan ROS dapat dihambat dengan pemberian antioksidan salah satunya antosianin yang merupakan salah satu kandungan dari ubi jalar ungu. Pada penelitian ini ekstrak ubi jalar ungu dibagi menjadi dua konsentrasi yaitu 10% dan 15% yang diberikan secara topikal pada luka tikus, kemudian dihitung persentase penutupan lukanya pada hari ke 3, 7, dan 14 setelah pembuatan luka. Hasil uji One Way Anova menunjukkan bahwa ekstrak ubi jalar ungu memiliki hasil persentase penutupan luka yang signifikan pada hari ke-3 dengan nilai signifikansi (p = 0,000) < 0,05, sedangkan pada hari ke-7 dan ke-14 tidak menunjukkan hasil yang signifikan dan ekstrak ubi jalar ungu konsentrasi 15% memiliki persentase penutupan luka lebih besar dibandingkan dengan ekstrak konsentrasi 10%. Sehinggga dapat disimpulkan bahwa pemberian ekstrak ubi jalar ungu memiliki pengaruh terhadap penutupan luka yang signifikan pada hari ke-3 penyembuhan luka dan konsentrasi 15% masih merupakan konsentrasi terapeutik pada ekstrak ubi jalar topikal.

English Abstract

Diabetes mellitus is a chronic disease caused by deficiency of insulin production or defective insulin application that can occur due to inherited and acquired factors. One of the complication caused by uncontrolled diabetes is diabetic ulcer. The increase of Reactive Oxygen Species (ROS) causes disruption in the blood vessel system which is peripheral blood vessels and nerves which eventually leads to diabetic ulcer. Increased ROS from the endothelial healing process of wound in diabetes also cause decrease in growth factors TGF-β, so that decrease synthesis of collagen, epitelisation, and angiogenesis which causes slow wound healing. The purpose of this research is to know the effects of purple sweet potato extract topically in wound closure percentage on Rattus norvegicus strain wistar induced diabetes. Increase of ROS can be done by administering antioxidant such as anthocyanin which is one of the active constituent of purple sweet potato (Ipomoea batatas L.). The purple sweet potato extract is divided into two concentration of 10% and 15% respectively given topically on wound on rats, the the percentage of wound closure is calculated on 3rd, 7th, and 14th day after making the wound. Based on the research results One Way Anova test, indicated that the extract of purple sweet potato has significant result on wound closure on 3rd day with the significant value of (p = 0.000) < 0.05. Whereas on the 7th and 14th day showed no significant results and the purple sweet potato extract concentration of 15% has wound closure percentage greater than concentration of and the purple sweet potato extract concentration of 15% have a percentage of closing wounds greater than concentration of 10%. So, it can be concluded that the administration of the purple sweet potato extract topically has significant effects on the 3rd day of the wound healing process in diabetic rat models and the concentration of 15% is still a therapeutic concentration in the sweet potato extract topically
