Abstract
Hiperkolesterolemia adalah gangguan metabolisme kolesterol yang disebabkan oleh meningkatkan kadar kolesterol yang melebihi batas normal. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian ekstrak jintan hitam (Nigella sativa) terhadap kadar trigliserida (TG) dan gambaran histologi tikus model hiperkolesterolemia. Penelitian ini menggunakan hewan model tikus (Rattus Norvegicus) jantan strain wistar dengan berat badan kisaran 150-200 g. Induksi hiperkolesterolemia menggunakan diet pakan tinggi lemak (asam kolat 0,02 g, minyak babi 2g dan kuning telur puyuh rebus 1 g) kgBB selama 28 hari berturut-turut. Penelitian ini bersifat eksperimental dengan menggunakan 5 kelompok tikus dengan 4 ulangan yaitu kelompok kontrol positif, kontrol negatif, dan 3 kelompok terapi dengan dosis 150 mg//kgBB (T1), 300 mg/kgBB (T2), dan 450 mg/kgBB (T3) selama 14 hari dengan pemberian secara oral. Kadar trigliserida (TG) diukur dengan metode GPO-PAP dan gambaran histopatologi jantung dengan pewarnaan Hematoksilin-Eosin (HE). Data dianalisis secara kualitatif dan kuantitatif. Hasil menunjukan bahwa ektrak jintan hitam (Nigella sativa) secara signifikan (p<0,05) menurunkan kadar trigliserida (TG) dan memperbaiki gambaran histopatologi jantung tikus model hiperkolesterolemia dengan dosis terbaik 450 mg/BB.
English Abstract
Hypercholesterolemia is a metabolism disruption of cholesterol caused by the increased levels of cholesterols that exceed the normal level. The purpose of this study is to find out the effect of black caraway (Nigella sativa) extract on triglycerides (TG) level and histopathology appearance of heart in hypercholesterolemia rats. This study used male Wistar rats (Rattus norvegicus) as the model animals, weigh around 150-200 grams. Hypercholesterol induction used high fat food diet (0,02 gram cholic acid, 2 gram lard, and 1 gram boiled quail egg yolk)/kg BW for 28 days. This study was experimental that used five groups of rats and four rats for each group of : positive group, negative group, and three groups of therapy with a dose of 150 mg/kg BW (T1), 300 mg/kg BW (T2), and 450 mg/kg BW (T3) for 14 days, with orally administered. Triglycerides (TG) level were analyzed using GPO-PAP and heart histopathology using Hematoxyline Eosin (HE) staining. Data were analyzed qualitative and quantitative. The result showed that extract black caraway (Nigella sativa) significantly (p<0.05) decrease triglycerides level and repair histopathology of heart on hypercholesterolemia rats was the best dose of 450 mg/kg BW.
