Abstract

Gastroenteritis adalah suatu inflamasi pada organ lambung dan intestine yang ditandai dengan gejala muntah dan diare. Penyebab gastroenteritis salah satunya adalah bakteri Escherichia coli. Green coffee Robusta (Coffea cannephora) mengandung CGA (chlorogenic acid) yang berfungsi sebagai antibakteri, antioksidan, dan antiinflamasi. Penelitian ini bertujuan untuk mengetahui ekstrak green coffee robusta dapat menurunkan ekspresi IL-1β dan memperbaiki histopatologi organ kolon tikus putih model gastroenteritis. Rancangan penelitian ini bersifat eksperimental menggunakan rancangan acak lengkap (RAL). Penelitian ini menggunakan tikus jantan strain wistar usia 8-12 minggu dengan berat badan 150-200gr. Pemberian dexamethason bertujuan untuk menurunkan sistem kekebalan tubuh terhadap tikus putih model gastroenteritis dengan dosis 5 mg/kgBB b.i.d secara intraperitoneal selama 3 hari. Infeksi E.coli diberikan menggunakan sonde lambung sebanyak 1 ml konsentrasi 3x108 selama 7 hari setelah pemberian dexamethasone. Tikus dibagi menjadi 5 kelompok yaitu terdiri dari kontrol negatif, kontrol positif, kelompok perlakuan diberi dexamethasone dosis 5 mg/kgBB dan ekstrak kopi unroasted robusta dengan dosis 500 mg/kgBB, 1.000 mg/kgBB, dan 1.500 mg/kgBB. Ekspresi IL-1β organ kolon menggunakan metode imunohistokimia, pengukuran menggunakan immunoratio dianalisa dengan one way ANOVA dengan tingkat kepercayaan α = 0,05 dan histopatologi organ kolon dianalisa secara deskriptif. Hasil penelitian menunjukkan pemberian preventif ekstrak green coffee robusta dengan dosis 1500mg/kgBB mampu menurunkan ekspresi IL-1β dan dapat memperbaiki kerusakan organ kolon yang ditandai dengan tidak adanya erosi epitel pada lapisan mukosa, jumlah sel goblet dan sel radang normal.

English Abstract

Gastroenteritis is an inflammation that occurs in stomach and intestines which characterized by symptoms like vomiting and diarrhea. One of the causes of gastroenteritis is Escherichia coli. Green coffee Robusta (Coffea cannephora) consists of CGA (chlorogenic acid) that serves as antibacterial, antioxidant and anti-inflammation. The purpose of this study was to investigate the effect of green coffee robusta in reducing IL-1β expression and colon histopathological image of white rats (Rattus norvegicus) gastroenteritis model. The design of this study was experimental and used Completely Randomized Design (CRD). This study used 20 male wistar-strain rats age 8-12 weeks and weight between 150-200g. The administration of dexamethasone intends to reduce the immune system of white rats gastroenteritis model with dose of 5 mg/kg BW b.i.d via intraperitoneal injection for 3 days. Infection of 1 mL E.coli 3x108 used gavage for 7 days after the administration of dexamethasone. The rats were divided into 5 groups consist of negative control, positive control, treatment groups. Treatment groups (P1, P2, P3) those received green coffee extract with dosage of 500 mg/kg BW, 1.000 mg/kg BW and 1.500 mg/kg BW respectively prior to receive of Dexamethasone and E.coli. Expression of colon IL-1β measured using immunohistochemistry method and the data were analyzed by one-way ANOVA with confidence level of α = 0,05. Colon histopathology image were analyzed descriptively. The results of this study showed that preventive of green coffee robusta extract with best dose of 1.500 mg/kg BW was able to reduce the expression of IL-1β and could repair colonic organ damage which was characterized by the absence of epithelial erosion in the mucosal layer and inflammatory cells. In addition the number of goblet cells show normal.
