Abstract

Pneumonia termasuk salah satu penyebab terbesar mortalitas, terutama pada anak balita dan orang tua. Streptococcus pneumoniae merupakan patogen yang paling sering menyebabkan pneumonia komuniti atau pneumonia yang didapat dari lingkungan. Terapi pneumonia saat ini adalah β – lactam, macrolide generasi baru, fluoroquinolone. Mengingat adanya kemungkinan resistensi terhadap antibiotik, perlu dikembangkan antibiotik baru terhadap Streptococcus pneumoniae. Mahkota dewa yang seringkali menjadi taman hias kini mulai banyak digunakan untuk pengobatan. Beberapa penelitian yang telah dilakukan membuktikan bahwa terdapat zat aktif seperti flavonoid, tanin, saponin. Flavonoid buah mahkota dewa dapat digunakan sebagai antimikroba pada bakteri lain, tetapi efek flavonoid buah mahkota dewa terhadap Streptococcus pneumoniae belum pernah diteliti. Penelitian ini bertujuan untuk mengetahui potensi antimikroba flavonoid ekstrak buah mahkota dewa (Phaleria macrocarpa) terhadap Streptococcus pneumoniae dengan menggunakan desain eksperimentaI laboratorium melalui uji potensi antimikroba metode difusi sumuran. Konsentrasi flavonoid ekstrak buah mahkota dewa yang digunakan adalah 10%, 20%, 40%, 60%, 80%, 100% dengan empat kali pengulangan. Berdasarkan hasiI peneIitian, zona inhibisi terbentuk mulai konsentrasi 20%. Analisa statistik menggunakan One-Way ANOVA Test menunjukkan perbedaan yang signifikan pada perubahan konsentrasi flavonoid ekstrak buah mahkota dewa terhadap zona inhibisi pertumbuhan bakteri Streptococcus pneumoniae (p<0.05). Uji korelasi Pearson menunjukkan adanya hubungan yang sangat kuat dengan arah positif sehingga dapat disimpulkan semakin meningkatnya konsentrasi flavonoid ekstrak buah mahkota dewa, maka potensi antimikroba semakin kuat. Berdasarkan penelitian ini, dapat disimpulkan bahwa flavonoid ekstrak buah mahkota dewa memilikipotensi sebagai antimikroba terhadap bakteri Streptococcus pneumoniae secara in vitro.

English Abstract

Pneumonia is one of the major causes of mortality, especially for children and geriatry. Most common causes of community acquired pneumoniae (CAP) is Streptococcus pneumoniae. Nowadays, the drug of choice for pneumoniae are β – lactame, new macrolide, and fluoroquinolone. Because of the possibility of antimicrobial resistance, this kind of experiment is needed for discovering a new antimicrobial for Streptococcus pneumoniae. People were used Phaleria macrocarpa for decorative plants, but now, Phaleria macrocarpa is common use for treatment. There are so many research about the bioactive component in Phaleria macrocarpa, such as flavonoid, tanin, saponin that considered to play a role in antimicrobial properties. The flavonoid can use for antimicrobial for another bactery, but there are no research for its efficacy for Streptococcus pneumoniae. This research was a laboratory experiment design using well-diffusion method, aimed to know the antimicrobial potention of flavonoid of extract of Phaleria macrocarpa against Streptococcus pneumoniae. The concentration of flavonoid was 100%, 80%, 60%, 40%, 20%, 10%, and 0% with four repetitions. Inhibition zones was observed in all concentration. Statistical analysis using One-Way ANOVA test showed a significant difference in the consentration changes toward Streptococcus pneumoniae inhibition zone of bacterial growth (p <0.05). Pearson correlation test showed a very strong relationship with positive direction which can be interpretated that the higher concentration of flavonoid, the stronger antimicrobial potential. Based on this in vitro research, it can be concluded that flavonoid of Phaleria macrocarpa extract has an antimicrobial potential toward Streptococcus pneumoniae.
