Abstract

Stroke iskemik merupakan suatu penyakit defisit neurologis yang disebabkan oleh gangguan pembuluh darah otak dan dapat menyebabkan mortalitas dan morbiditas yang tinggi . stroke iskemik menyebabkan nekrosis sel pada otak dengan salah satu jalur mekanismenya melalui induksi kaskade glutamate secara massif dan tidak terkontrol. Induksi glutamate secara massif ini dapat memperluas dan memperparah kerusakan yang diakibatkan stroke iskemik. Saccharomyces cerevisiae adalah khamir uniseluler yang merupakan galur potensial penghasil Beta-glukanyang berpotensi mengambat kaskade glutama tdan berdiferensiasi menjadi neuron sehingga dapat mengembalikan struktur dan fungsional otak. Tujuan dari penelitian ini adalah membuktikan potensi ekstrak beta-glukandari S. cerevisiae dalam menghambat distribusi glutamate dan memperbaiki histopatologi jaringan otak pada tikus model stroke iskemik. Penelitian ini adalah eksperimen murni dengan metode Randomized Post Test Only Controlled Group Design. Penelitian ini dilakukan secara in vivo menggunakan tikus model stroke iskemik yang dibagi menjadi 5 kelompok yaitu kontrol negatif, kontrol positif, perlakuan 1, perlakuan 2, dan perlakuan 3. Kelompok kontrol positif dan perlakuan diinduksi stroke iskemik dengan metode Middle Cerebral Artery Occlusion (MCAO). Kelompok perlakuan 1, 2 dan 3 diterapi dengan larutan Beta-glukandari ekstrak Saccaromycesses cerevisae 18, 36, 72 mg/kgBB setelah penginduksian selama 2 minggu. Tikus dibedah untuk pemeriksaan Ekspresi Glutamat dan pembuatan histopatologi jaringan darah pada minggu pertama dan kedua setelah terapi dilaksanakan. Hasil pengukuran Immunohistokimia untuk pengecatan glutamat diharapakan Setelah Kaskade glutamat dihambat akan menurunkan angka kerusakan yang diakibatkan oleh stroke iskemik . Dengan demikian, beta-glukanberpotensi menjadi terapi stroke iskemik yang efektif

English Abstract

Ischemic stroke is a neurological deficit disease caused by a brain blood vessel disorder and can cause high mortality and morbidity. Ischemic stroke causes cell necrosis in the brain with one of its pathways through massive and uncontrolled induction of the glutamate cascade. This massive glutamate induction can expand and worsen the damage caused by ischemic stroke. Saccharomyces cerevisiae is a unicellular yeast which is a potential Beta-glucan-producing strain that has the potential to inhibit glutamate cascade and differentiate into neurons so that it can repair brain structure and function. The purpose of this study is to prove the potential of betaglucan extract of S. cerevisiae in inhibiting glutamate distribution and improving histopathology of brain tissue in ischemic stroke mice. This research is a pure experiment with the Randomized Test Only Controlled Group Design method. This study was conducted in vivo using ischemic stroke models divided into 5 groups: negative control, positive control, treatment 1, treatment 2, and treatment 3. Positive control group and ischemic stroke-induced treatment with Middle Cerebral Artery Occlusion (MCAO) method. Treatment groups 1, 2 and 3 were treated with Beta-glucan extract from Saccaromycesses cerevisae 18, 36, 72 mg / kgBW after induction for 2 weeks. Mice were dissected for examination of glutamate expression and histopathology of blood tissue in the first and second weeks after therapy was carried out. The results of immunohistochemical measurements for glutamate staining are expected after the glutamate cascade is inhibited will reduce the number of damage caused by ischemic stroke. Thus, betaglucan has the potential to be an effective ischemic stroke therapy.
