Abstract
Hiperkolesterolemia adalah suatu keadaan tingginya kadar kolesterol dalam darah. Peningkatan metabolisme lipid menyebabkan timbulnya peroksidasi lipid yang mengakibatkan kerusakan sel. Ekstrak jinten hitam memiliki kandungan saponin, niasin dan thymoquinone yang berfungsi membantu menurunkan kolesterol berlebih didalam tubuh serta sebagai antioksidan untuk mencegah terjadinya peroksidasi lipid. Tujuan penelitian ini adalah untuk mengetahui pengaruh ekstrak jinten hitam sebagai terapi hiperkolesterolemia ditinjau dari kadar Malondialdehid (MDA) dan gambaran histopatologi jejunum. Penelitian ini menggunakan 20 ekor tikus putih strain Wistar berumur 10-12 minggu dengan berat badan 150-200 gr yang kemudian dibagi menjadi kelompok kontrol negatif, kelompok kontrol positif (serta kelompok pemberian terapi ekstrak jinten hitam dosis P1 150 mg/kg BB, P2 300 mg/kg BB dan P3 450 mg/kg BB. Diet tinggi lemak diberikan selama 28 hari, dilanjutkan dengan pemberian ekstrak jinten hitam dan diet hiperkolesterol secara bersamaan selama 14 hari. Pengukuran kadar MDA menggunakan metode uji Thiobarbituric Acid (TBA) dan dianalisa secara kuantitatif menggunakan one way ANOVA yang dilanjutkan uji Tukey dengan tingkat kepercayaan 95% (α =0,05). Pengamatan histopatologi dilakukan dengan pewarnaan Hematoksilin Eosin (HE) diamati menggunakan mikroskop dan dianalisa secara kualitatif deskriptif. Hasil penelitian menunjukkan bahwa dosis terapi 450 mg/kgBB menurunkan kadar MDA pada sebesar 30,20% dan menunjukkan perbaikan gambaran histopatologi tunika mukosa jejunum tikus hiperkolesterolemia. Kesimpulan dari penelitian ini menunjukkan bahwa ekstrak jinten hitam (Nigella sativa) dapat sebagai terapi yang ditunjukkan dengan menurunkan kadar MDA dan memperbaiki gambaran histopatologi tunika mukosa jejunum tikus hiperkolesterolemia.
English Abstract
Hypercholesterolemia is a condition where the cholesterol level in blood increased. The increase of lipid metabolism leads to lipid peroxidation that eventually will lead to the damage of cells. Black cumin extract contains saponin, niacin and thymoquinone which functions to help reduce excess cholesterol in the body and as an antioxidant to prevent the occurrence of lipid peroxidation. The purpose of this study was to find out the effect of black cumin extract as hypercholesterolemia therapy based on Malondialdehyde (MDA) level and histopathology image of jejunum. This study used 20 Wistar white rats age 10-12 weeks and weighed 150-200 gr that were divided into negative control group, positive control group and the black cumin extract therapy group with doses of P1 150 mg/kgBW, P2 300 mg/kgBW and P3 450 mg/kgBW. High fat diet was given for 28 days, continued by the administration of black cumin extract and hypercholesterol diet simultaneously for 14 days. The MDA level was measured using Thiobarbituric Acid (TBA) test method and was analyzed quantitatively using one-way ANOVA, continued by Tukey test with confidence level of 95% (α =0,05). The histopathology observation was conducted with Hematoksilin Eosin (HE) staining, and was observed under the microscope and was analyzed qualitatively and descriptively. The result of this study showed that the therapy dose of 450 mg/kgBW lower the MDA level by 30,20% and showed an improvement in histopathology image of jejunum tunica mucosa of white rats with hypercholesterolemia. The conclusion of this study showed that black cumin extract (Nigella sativa) could be used as the therapy showed by the decreased level of MDA and improvement of histopathology image of jejunum tunica mucosa of white rats with hypercholesterolemia.
