Abstract

Diet aterogenik memicu peningkatan kadar kolesterol plasma yang berakibat pada penimbunan LDL (Low Density Lipoprotein) pada lapisan tunica intima dan tunica media aorta sehingga memicu proses aterosklerosis. Penimbunan LDL pada lapisan arteri akan memicu peningkatan ROS (Reactive Oxygen Species) yang beakibat pada timbulnya respon inflamasi vaskuler. Respon Inflamasi vaskuler diperankan oleh sitokin inflamasi TNF-α yang dihasilkan oleh makrofag pada saat proses oksidasi LDL. Pencegahan aterosklerosis dapat dilakukan dengan agen antiinflamasi, salah satu agen inflamasi adalah fucoidan yang terdapat pada ekstrak etanol Sargassum sp. Penelitian eksperomental laboratorik ini bertujuan mengetahui pengaruh ekstrak etanol Sargassum sp terhadap ekspresi TNF-α aorta pada tikus Rattus norvegicus strain Wistar yang diinduksi diet aterogenik. Metode penelitian adalah Randomized Post Test Only Controlled Group Design dengan menggunakan 30 ekor tikus. Tikus dibagi menjadi 6 kelompok yakni kontrol negatif, kontrol positif, perlakuan dosis 1 (150 mg/kgBB), dosis 2 (300 mg/kgBB), dosis 3 (600 mg/kgBB), dan simvastain (4 mg/kgBB). Uji One-Way ANOVA menunjukkan perbedaan nilai yang signifikan (p<0,05) pada pemberian ekstrak etanol Sargassum sp pada tikus yang diinduksi diet aterogenik. Pemberian dosis Sargassum sp 150 mg/KgBB mampu menurunkan ekspresi TNF-α aorta yang signifikan (p<0,05) dibanding kelompok dosis lain. Sehingga dapat disimpulkan bahwa pemberian ekstrak etanol Sargassum sp pada tikus yang diinduksi diet aterogenik dapat menurunkan ekspresi TNF-α.

English Abstract

Atherogenic diet triggers an increase in plasma cholesterol levels which results in the accumulation of LDL in tunica intima and tunica media layers which stimulates the atherosclerosis process. The mass of LDL in the arterial layers will cause ROS increase which results in the emergence of a vascular inflammatory response. The trigger of vascular inflammatory response is inflammatory cytokines TNF-α produced by macrophages during the LDL oxidation process. However, it is possible to prevent the atherosclerosis process by utilizing antiinflammation agents such as fucoidan found in the ethanolic extract of Sargassum sp. This experimental laboratory study aims to determine the effect of ethanolic extract of Sargassum sp on TNF-α aorta expression in Rattus norvegicus induced by atherogenic diets. The research method used is Randomized Post Test Only Controlled Group Design using 30 rats which consisted of 6 groups: negative control, positive control, dose 1 (150 mg / kg body weight), dose 2 (300 mg / kg body weight), dose 3 (600 mg / kg body weight), and simvastatin (4 mg / kg body weight). The One-Way ANOVA test showed that the administration of ethanolic extract of Sargassum sp in atherogenic diet-induced rats could reduce aortic TNF-α expression significantly (p <0.05). Furthermore, the dosage of this extract which had the best effect was 150 mg / Kg body weight (p <0.05).
