Abstract

Ovariektomi yaitu pengambilan ovarium pada hewan betina sehingga ovarium tidak dapat menghasilkan estrogen. Penurunan hormon estrogen mengakibatkan penurunan absorbsi kalsium di usus dan peningkatan hormon paratiroid dalam meresorpsi kalsium di tulang dan meningkatkan ekspresi IL-6 dapat mengakibatkan osteoporosis. Pencegahan osteoporosis dengan cara pemberian asupan kalsium dari luar tubuh dengan tepung cangkang udang vannamei (Litopenaeus vannamei). Tujuan penelitian untuk mengetahui pengaruh tepung cangkang udang vannamei terhadap kadar estrogen dan ekspresi IL-6 pada tikus putih (Rattus norvegicus) model ovariektomi. Tikus terbagi dalam 5 kelompok yaitu kelompok kontrol positif, kelompok kontrol negatif, kelompok terapi 500 mg/kgBB, kelompok terapi 1000 mg/kgBB, kelompok terapi 1500 mg/kgBB. Pengukuran kadar estrogen dilakukan dengan metode ELISA dan ekspresi IL-6 dilakukan dengan metode imunohistokimia. Analisa data dilakukan secara kuantitatif menggunakan analisa ragam ANOVA dan uji lanjutan Tukey α = 5%. Hasil penelitian menunjukan bahwa pemberian tepung cangkang udang meningkatkan kadar estrogen pada terapi 500 mg/KgBB sebesar 43%, dosis 1000 mg/KgBB sebesar 85%, dan dosis 1500 mg/KgBB sebesar 56%. Selain itu pemberian tepung cangkang udang juga berpengaruh dalam menurunkan ekspresi IL-6 pada dosis 500 mg/KgBB sebesar 92%, dosis 1000 mg/KgBB sebesar 62%, dosis 1500 mg/KgBB sebesar 70% secara signifikan (p<0,05). Disimpulkan bahwa pemberian tepung cangkang udang vannamei dosis 1000 mg/KgBB pada tikus putih (Rattus norvegicus) model ovariektomi dapat meningkatkan kadar hormon estrogen sebanyak 85% dan menurunkan ekspresi IL-6 sebanyak 92%.

English Abstract

Osteoporosis is one of the effects of ovariectomy treatment, namely the removal of ovaries in female animals so that the ovaries can not produce estrogen. Decreased estrogen hormone resulted in decreased calcium absorption in the intestine and increased parathyroid hormone in the calcium receptor in bone and increased expression of IL-6 cytokines. Prevention of osteoporosis is done by providing calcium intake from outside the body with shrimp vannamei shrimp (Litopenaeus vannamei). The purpose of this research is to know the effect of shrimp vannamei shrimp on estrogen level and IL-6 expression on white rat (Rattus norvegicus) model of ovariectomy. Rats were divided into 5 groups: positive control group, negative control group, 500 mg / kgBB, mice group of 1000 mg / kgBB, mice group 1500 mg / kgBW. Measurement of estrogen levels was done by ELISA method and IL-6 expression was performed by immunohistochemical method. Data analysis was done quantitatively using ANOVA variance analysis and continued Tukey test α = 5%. The results showed that shrimp shell powder increased estrogen level significantly (P <0.05) dose 500 mg / KgBB by 43%, dose 1000 mg / KgBB by 85%, and dose 1500 mg / KgBB by 56%. In addition, shrimp shell powder was also significantly (p <0.05) in decreasing the expression of IL-6 at doses of 500 mg / KgBB by 92%, dose 1000 mg / KgBB by 62%, dose 1500 mg / KgBB by 70%. It was concluded that dosage shrimp vannamei dosage 1000 mg / KgBB in white rat (Rattus norvegicus) model of ovariectomy can increase estrogen hormone level as much as 85% and decrease expression of IL-6 as much as 92%.
