Abstract

Hiperglikemia kronis dapat menimbulkan kerusakan organ tubuh, khususnya ginjal. Kerusakan organ ginjal akibat hiperglikemia menjadi salah satu faktor resiko terjadinya urolithiasis. Salah satu pencegahan yang dapat dilakukan yaitu dengan memberikan tanaman semanggi air. Semanggi air (Marsilea crenata) memiliki kandungan kalium dan flavonoid yang berperan sebagai peluruh kristal, antihiperglikemia, antioksidan, dan antiinflamasi. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian perasan daun dan tangkai semanggi air untuk mencegah urolithiasis pada hewan model tikus (Rattus novergicus) hiperglikemia ditinjau dari kadar TNF-α dan IL-1β. Penelitian ini menggunakan RAL (Rancangan Acak Lengkap) dengan metode eksperimental. Kelompok perlakuan dibagi menjadi 5, yaitu P1 (kontrol positif urolithiasis), P2 (kontrol positif hiperglikemia dan urolithiasis), dan pemberian perasan daun dan tangkai semanggi air dibagi menjadi 3 konsentrasi yaitu P3 (20%), P4 (40%), dan P5 (60%). Variabel penelitian berupa kadar TNF-α dan IL-1β dengan metode indirect Elisa yang dianalisis statistika dengan uji one-way ANOVA dan uji lanjutan (Posthoc Test) dengan taraf kepercayaan 95% (α=0,05). Hasil penelitian menunjukkan pemberian perasan daun dan tangkai semanggi air secara signifikan dapat mencegah kenaikan kadar TNF-α dan IL-1β pada konsentrasi yang paling baik sebesar 60%. Disimpulkan bahwa perasan daun dan tangkai semanggi memiliki potensi sebagai prevensi urolithiasis pada hewan model hiperglikemia melalui penurunan kadar TNF-α dan IL-1β

English Abstract

Chronic hyperglycemia can cause damage to organs, especially the kidneys. Kidney organ damaged due to hyperglycemia might be one of the factors of risk occurrence of urolithiasis. One of the prevention that can be conducted is to give Marsilea crenata. Marsilea crenata contains potassium and flavonoid which all of those compound acted as set crystals, antihyperglycemic, antioxidant, and anti-inflammatory. This study was aimed to know the influence of the giving Marsilea crenata leaves and stalks juice in preventing the occurrence of urolithiasis in hyperglycemia rats (Rattus novergicus) in toward of TNF-α and IL- 1β. Treatment is divided into 5 groups, namely P1 (positive control urolithiasis), P2 (positive control hyperglycemia and urolithiasis), and P3, P4, P5 (induction hyperglycemia and Marsilea crenata leaves and stalks juice divided into several concentration i.e. 20%, 40%, 60% with induction urolithiasis. The study variable was TNF-α and IL-1β level with Elisa indirect method was analyzed statistically with one-way ANOVA test and advanced test (Posthoc Test) with 95% (α = 0,05) confidence level. The result showed Marsilea crenata leaves and stalks juice can prevent the increase in level of TNF-α and IL-1β at the best concentration of 60%. It was concluded that Marsilea crenata leaves and stalks juice have effect as prevention of urolithiasis in animal models of hyperglycemia through the decrease in level of TNFα and IL-1β.
