Abstract

Porphyromonas gingivalis (P.g) merupakan bakteri anaerob gram negatif dalam rongga mulut yang berperan dalam pathogenesis terjadinya periodontitis. Periodontitis merupakan kondisi inflamasi kronis dari periodontium yang dapat disebabkan oleh beberapa jalur patogenesis, salah satunya melalui radikal bebas dan stress oksidatif yang dapat menciptakan Reactive Oxidative Stress (ROS). Total katekin memiliki kandungan antioksidan yang dapat menurunkan kadar radikal bebas di dalam tubuh. Penelitian ini bertujuan untuk mengetahui pengaruh total katekin terhadap kadar GSH (Glutathione) pada model periodontitis tikus wistar jantan (rattus norvegicus) yang diindksi bakteri Pophyromonas gingivalis. Penelitian ini menggunakan True Experimental Single Blinded Design yaitu Postest Only Control Group Design. Kelompok kontrol terdiri dari dua kelompok, yaitu kelompok kontrol negatif adalah hewan coba yang tidak diberi perlakuan apapun dan kelompok kontrol positif adalah hewan coba yang diberi perlakuan injeksi bakteri Pg ATCC 33277 selama 28 hari, kelompok perlakuan terdiri dari 3 kelompok yaitu hewan coba yang diinjeksi bakteri Pg ATCC 33277 selama 28 hari dan diberi penyondean total katekin dengan dosis yang berbeda yakni 100mg/kgBB, 200 mg/kgBB dan 400 mg/kgBB. Hasil penelitian menunjukkan bahwa tidak terdapat pengaruh total katekin yang signifikan pada kadar GSH. Kesimpulan dari penelitian ini adalah total katekin pada dosis 400 mg/kgBB berpengaruh signifikan terhadap kadar GSH model periodontitis.

English Abstract

Porphyromonas gingivalis (P.g) is gram negative bacteria in oral cavity that play role in pathogenesis of periodontitis. Periodontitis is chronic inflammatory condition from periodontium that cause by several way of pathogenesis, one of them is free radical and oxidative stress which produce Reactive Oxidatives Stress (ROS), so it can leading to tissue damage through lipid peroxidation in a tissue, DNA and protein damage, enzyme oxidation, stimulation of pro-inflammatory cytokines. Green tea is a plant that contain fluoride, catechin and polyphenol. Total catecin also contain antioxidant which can reduce free radical in the body. The purpose of this experiment is to know the effect of total catechin to GSH level in periodontitis’ model rats (rattus norvegicus) that induced by Pophyromonas gingivalis. This study used a True Experimental Single Blinded Design, Postest Only Control Group Design. The control group divided in two group, negative group control is the group which not give any induction and treatment and positive group is the group which indued by bacteri Pg ATCC 33277 until 28 days, experiment group is divided in 3 group, it is given induced by bacteria Pg ATCC 33277 until 28 days and receive gauge of total catechin with different dose, there are dose 100 mg/kgBB, 200 mg/kgBB and 400 mg/kgBB. The result of this study show there are not significant effect of total catechin to GSH concentration Conclusion of this study is total catchin just give significant result to GSH concentration in dose 400 mg/kgBB.
