Abstract
Diabetes Melitus (DM) merupakan sekumpulan penyakit metabolik yang ditandai dengan hiperglikemia sebagai akibat dari defek sekresi insulin, kerja insulin, atau keduanya. Hiperglikemia kronis pada DM diasosiasikan dengan terjadinya penurunan fungsi kognitif dan memori pada penderitanya yang diwakili dengan meningkatnya kehilangan neuron pada kortex cerebri. Hiperglikemia memicu produksi advanced glycation end-products (AGEs) dan merusak sel-sel target melalui beberapa mekanisme. AGEs memodifikasi protein intraseluler sehingga fungsinya menjadi berubah, memicu produksi reactive oxygen species (ROS), dan meningkatkan ekspresi gen-gen pro-inflamasi. Vitamin A bersifat antioksidan karena struktur rantai hidrofobik unit polietilen yang dapat menetralkan radikal bebas. Tujuan dari penelitian ini adalah untuk membandingkan pengaruh pemberian vitamin A terhadap persentase sel apoptosis terhadap sel normal lapisan neuron piramidal pada tikus (Rattus norvegicus) galur Wistar jantan model diabetes melitus tipe 2. Penelitian ini menggunakan uji eksperimental murni secara in vivo di laboratorium dengan rancangan Randomized Post Test Only Controlled Group Design. Sampel histopatologi diamati dengan pengecatan HE di bawah perbesaran objektif 40X. Hasil analisis menggunakan one-way ANOVA diperoleh nilai signifikansi 0.003 dan koefisien korelasi Spearman diperoleh (r) -0.791 yang berarti korelasi negatif yang kuat. Kesimpulan pada penelitian ini yaitu terdapat pengaruh signifikan dari pemberian vitamin A terhadap berkurangnya persentase sel apoptosis terhadap sel normal neuron piramidal pada tikus model diabetes melitus tipe 2.
English Abstract
Diabetes Mellitus (DM) is a cluster of metabolic diseases characterized by hyperglycaemia resulted from insulin secretion defect, insulin activity defect or both. Chronic hyperglycaemia in DM is associated with memory and cognitive function impairment that represented by the increase in neuronal loss in cerebral cortex. Hyperglycaemia triggers production of advanced glycation end-products (AGEs) and disrupts target cells through several pathways. AGEs modify intracellular proteins and impair their function, lead to reactive oxygen species (ROS) production, and promote expression of pro-inflammatory genes. Vitamin A has antioxidant property because of its chains of hydrophobic polyethylene unit structure that neutralize free radicals. The objective of this experiment is to compare the effect of Vitamin A on Percentage between Apoptosis and Normal Pyramidal Neurons in Cerebral Cortex of Type 2 Diabetic Rats Model. This study is conducted with pure experimental design in vivo in the laboratory with Randomized Post Test Only Controlled Group. Histological samples are observed with H&E staining under objective magnification of 40X. The data is analysed with one-way ANOVA with significance value of 0.003 and Spearman correlation coefficient of (r) -0.791 which means strong negative correlation. It is concluded that there is significant effect of vitamin A on percentage between apoptosis and normal pyramidal neuron in cerebral cortex of type 2 diabetic rats model
