Abstract
Toxoplasma gondii adalah salah satu protozoa penyebab infeksi kronis yang diduga menyebabkan obese (infectobesity). Profilin merupakan salah satu komponen di dalam alat gerak Toxoplasma gondi yang dapat meningkatkan interleukin-12 (IL-12) sehingga menyebabkan inflamasi adiposa dan diduga dapat menyebabkan obesitas dan sindroma metabolik. Sindroma metabolik dapat mengurangi sensitivitas reseptor insulin sehingga memengaruhi gula darah. Toxoplasma gondi juga dapat mempengaruhi kadar gula darah dengan merusak sel beta pankreas. Tujuan penelitian untuk mengetahui efek paparan profilin Toxoplasma gondii terhadap kadar gula darah puasa (GDP) pada tikus diberi diet tingi kalori. Studi bersifat eksperimental menggunakan post-test only terhadap tikus Wistar jantan selama 16 minggu. Sampel terdiri dari 65 ekor tikus, terbagi 13 kelompok perlakuan. Variabel yang diukur adalah kadar GDP dengan menggunakan metode Trinder. Pengujian diagnostik menggunakan uji ANOVA, korelasi Pearson, dan uji regersi linear. Didapatkan hasil penelitian tidak ada perbedaan pengaruh dosis profilin terhadap kadar GDP (ANOVA p = 0,056). Hasil regresi linear pada kelompok injeksi satu kali yang diberi diet normal tidak signifikan, berkorelasi sangat lemah dan searah (p = 0,887; R = 0,075). Sedangkan pada kelompok injeksi satu kali yang diberi diet tinggi kalori hasilnya tidak signifikan, berkorelasi lemah dan searah (p=0,195; R=0,342). Kelompok injeksi dua kali yang diberi diet normal hasilnya tidak signifikan, berkorelasi lemah dan berlawanan arah (p=0,472; R=-0,209). Hasil uji pada kelompok injeksi dua kali yang diberi diet tinggi kalori tidak signifikan, tidak berkorelasi dan berlawanan arah (p=0,901; R=- 0,037) Delta berat badan tikus memiliki perbedaan pengaruh terhadap dosis profilin Toxoplasma gondii (ANOVA p=0,000). Dapat disimpulkan bahwa paparan profilin Toxoplasma gondii tidak memengaruhi kadar GDP.
English Abstract
Toxoplasma gondii is one of the protozoan causes of chronic infection that allegedly causes obese (infectobesity). Profilin is one of the components in the Toxoplasma gondii toolkit that can promote interleukin-12 (IL-12), which causes adipose inflammation and is thought to cause obesity and metabolic syndrome. Metabolic syndrome can reduce the sensitivity of insulin receptors that affect blood sugar. Toxoplasma gondii can also affect blood sugar levels by damaging the pancreatic beta cells. The objective of the study was to investigate the effect of Toxoplasma gondii profilin dose on fasting blood glucose (FBG) level in mice given high calories diet. The experimental study used post-test only of male Wistar rats for 16 weeks. The sample consisted of 65 rats, divided into 13 treatment groups. The measured variable is the FBG content by using the Trinder method. Diagnostic testing using ANOVA assay, Pearson correlation, and linear regression test. The results showed no difference in the effect of dose profilin on FBG (ANOVA p = 0,056). The linear regression result in the one-time injection group given normal diet was not significant, correlated very weakly and unidirectional (p = 0.887; R = 0.075). While in the one-time injection group who were given a high-calories diet the results were not significant, correlated weakly and unidirectional (p = 0.195; R = 0.342). The double-injection group given a normal diet result was insignificant, correlated weakly and counterclockwise (p = 0.472; R = -0.209). The test results in the twotime injection group who were given a high-calories diet were insignificant, non-correlated and counterclockwise (p = 0.901; R = -0.037) The weight-loss rats had different effects on the dose of Toxoplasma gondii (ANOVA p = 0,000). It can be concluded that exposure to Toxoplasma gondii profilin does not affect FBG levels.
