Abstract

Tingginya angka kematian penderita Lupus Eritematosus Sistemik (LES) di Indonesia memacu para peneliti untuk berinovasi menemukan terapi alternatif sebagai pilihan pengobatan salah satu penyakit autoimun ini. Di samping efek samping akibat pengobatan konvensional jangka panjang, beberapa hasil terapi alternatif menggunakan agen biosimilar masih terlalu mahal bagi sebagian besar penderita LES. Tanaman cocor bebek (Bryophyllum pinnatum) yang kaya akan alkaloid, flavonoid, dan bryophillin A, bryophillin B, bryotoxin A, dan bryotoxin B yang mampu berikatan dengan reseptor BAFF dan APRIL yang berperan dalam maturasi sel B yang berpengaruh terhadap mekanisme autoantibodi pada LES. Penelitian ini bertujuan untuk mengetahui efek pemberian ekstrak daun cocor bebek terhadap fungsi ginjal mencit Balb/c model LES induksi pristan berdasarkan kadar ureum dan serum kreatinin. Metode penelitian yang digunakan adalah posttest only controlled group design menggunakan 25 ekor mencit. Mencit dibagi menjadi 5 kelompok yaitu kontrol negatif, kontrol positif, perlakuan dosis ekstrak daun cocor bebek P1 (10,5mg/kgBB/hari), P2 (21mg/kgBB/hari) dan P3 (42mg/kgBB/hari). Data dianalisa dengan uji normalitas (Shapiro-Wilk), uji homogenitas (Levene), dan uji ANOVA serta uji korelasi Pearson. Didapatkan perbedaan yang signifikan (p<0,05) antara kelompok P1 dengan P3 dan P2 dengan P3, meskipun perbedaan P1 dan P2 tidak terlalu signifikan (p>0,05). Pemberian ekstrak etanol daun Bryophyllum pinnatum pada dosis 10,5 ml/kgBB mampu menurunkan kadar ureum dan kreatinin serum pada mencit Balb/c model lupus induksi pristan.

English Abstract

The high mortality rate among systemic lupus erythematosus (SLE) patients in Indonesia has encourage researchers to find alternative therapies as a treatment option for this autoimmune disease. In addition to the side effects of the conventional therapy, alternative therapy results using biosimilar agents are still unaffordable for most patients. Bryophyllum pinnatum plant is rich in alkaloids, flavonoids, and bryophillin A, bryophillin B, bryotoxin A, and bryotoxin B which are able to bind to BAFF and APRIL receptors involved in B cell maturation lead to autoantibodi disorders in SLE. This study aims to determine the effects of Bryophyllum pinnatum leaves extract to Balb/c pristan-induced lupus mice renal function based on urea and serum creatinine levels. The research method used was posttest only controlled group design using 25 mice. Mice were divided into 5 groups: negative control, positive control, P1 given Bryophyllum pinnatum extract 10.5 mg/kgBW/day, P2 21 mg/kgBW/day and P3 42 mg/kgBW/day. Data were analyzed by normality test (Shapiro-Wilk), homogeneity test (Levene), ANOVA test, and Spearman test. There were significant differences (p <0.05) between P1 with P3 and P2 with P3, while the differences in P1 and P2 were not significant (p> 0.05).
