Abstract
Salah satu penyebab dari Nefropati diabetik (ND) yang paling berperan adalah peningkatan radikal bebas akibat tingginya kadar glukosa sehingga menyebabkan stress oksidatif. Stress oksidatif akan mengaktivasi Angiotensin II dan factor transkripsi Nuclear Factor kappa B (NFKb). Pencegahan dan perlambatan progresifitas ND adalah dengan menggunakan antioksidan. Salah satu contoh antioksidan adalah Vitamin E dan Asam Rosmarinat (AR). Vitamin E sebagai antioksidan konvesional sedangkan AR sebagai antioksidan poten yang juga memiliki efek antiinflamasi. Penelitian eksperimental laboratorik ini bertujuan membandingkan efek Vitamin E dan AR terhadap ekspresi NFKb p-65 pada glomelurus tikus diabetes tipe 2. Metode yang digunakan adalah randomized posttest only controlled group design dengan total tikus 15 ekor. Tikus dibagi kedalam 4 kelompok yaitu kontrol negatif, kontrol positif, perlakuan Vitamin E dan perlakuan AR. Data dianalisis dengan uji normalitas, uji homogenitas, uji One-way ANOVA, dan Post hoc test dengan LSD. Ekspresi NFKb p-65 kontrol positif meningkat signifikan dibandingkan kontrol negatif (p < 0,001), Vitamin E dan AR mampu menurunkan ekspresi NFKb dibanding dengan kontrol positif (p = 0,022 dan p = 0,001). Ekspresi NFKb pada kelompok Vitamin E dan Asam Rosmarinat tidak terdapat perbedaan signifikan namun AR menurunkan lebih baik dibandingkan Vitamin E. Dapat disimpulkan bahwa AR memberikan efek lebih baik pada penurunan ekspresi NFKb p-65 pada glomelurus tikus diabetes tipe 2 dibandingkan Vitamin E.
English Abstract
One of the causes of diabetic nephropathy (ND) that have important role is the increase of free radicals due to high levels of glucose which causes oxidative stress. Oxidative stress activates Angiotensin II and the transcription factor of Nuclear Factor kappa B (NFKb). Prevention and slowing down of ND progression is by using antioxidants. The example of antioxidants are Vitamin E and Rosmarinic Acid (RA). Vitamin E is a conventional antioxidant while RA is a potent antioxidant that also has anti-inflammatory effects. This laboratory experimental study aimed to compare the effects of Vitamin E and RA on the expression of NFKb p-65 in glomelurus of type 2 diabetes rats. The method used a randomized posttest only controlled group design with total of 15 rats. Rats were divided into 4 groups: negative control, positive control, Vitamin E, and RA treatments. Data were analyzed for normality, variant homogeneity, One-way ANOVA test, and Post hoc test using LSD. NFKb expression of p-65 of positive control increased significantly compared to negative controls (p <0.001), Vitamin E and RA were able to reduce NFKb expression compared to positive controls (p = 0.022 and p = 0.001). respectively there was no significant difference between NFKb expression in the Vitamin E group and Rosmarinic acid but RA decreased better than Vitamin E. It can be concluded that AR had a better effect compared to Vitamin E on the decrease in NFKb p-65 expression in glomelurus type 2 diabetes rats
