Uji Sitotoksik Kombinasi 5-Fluorourasil dan Coelomic Fluid dari Lumbricus rubellus Terhadap Cell Line HT-29 Secara In Vitro

Abstract

Kankerikolorektal merupakan salah satu jenis kanker yang terjadi pada mukosa kolon di mana penyakit ini mempunyai angka morbiditas dan mortalitas yang tinggi. kombinasi 5-fluorourasil dan coelomic fluid dari Lumbricus rubellus diharapkanXmenjadi salah satu alternatif terapi kombinasi yang mampu meningkatkan efek sitotoksik pada cell line HT-29. Uji sitotoksik dilakukan dengan metode uji MTT (3-(4,5-Dimethylthiazol-2-yl)-2,5-Diphenyltetrazolium Bromide) dengan pemberian konsentrasi kombinasi 5-fluorourasil dan coelomic fluid dari Lumbricus rubellus sebesar 5 µg/ml, 10 µg/ml dan 20 µg/ml. Kemudian hasil uji MTT dihitung menggunakan rumus persentase kematian sel. Dari hasil perhitungan persentase kematian sel didapatkan bahwa perlakuan 3 (20 µg/ml) dapat menghambat pertumbuhan sel paling tinggi. Kemudian data persentase kematian sel digunakan untuk mendapatkan nilai IC50 sebagai nilai penghambatan sebesar 50% terhadap perlakuan. Pada penelitian ini diperoleh nilai IC50 dari ratarata semua perlakuan sebesar 13,496 µg/ml. Data dari uji sitotoksik kombinasi 5- fluorourasil dan coelomic fluid dari Lumbricus rubellus terhadap cell line HT-29 diperiksaamenggunakan metode uji Kruskal Wallis. Pada uji Kruskal Wallis didapatkan data memiliki nilai signifikan (p < 0,05) sehingga data valid untuk digunakan sebagai data penelitian. Berdasarkan penelitianaini, didapatkan kesimpulan bahwa kombinasi 5-fluorourasil dan coelomic fluid dari Lumbricus rubellus mampu meningkatkan efek sitotoksik terhadap cell line HT-29 dengan konsentrasi coelomic fluid 20 µg/ml memiliki efek sitotoksik tertinggi dibanding semua perlakuan dan didapatkan nilai IC50 dari rerata semua perlakuan kombinasi 5-fluorourasil dan coelomic fluid dari Lumbricus rubellus sebesar 13,496 µg/ml.

English Abstract

Colorectal cancer is one type of cancer that occurs in the mucosa of the colon where the disease has a high morbidity and mortality. The combination of 5- fluourouracil and coelomic fluid from Lumbricus rubellus is expected to be one of the alternative combination therapy that can increase the cytotoxic effect on HT- 29 cell line. The cytotoxic test was performed by MTT assay (3-(4,5- Dimethylthiazol-2-yl)-2,5-Diphenyltetrazolium Bromide) by giving combination of 5- fluourouracil and coelomic fluid from Lumbricus rubellus on concetration 5 μg / ml, 10 μg / ml and 20 μg / ml. Then the MTT assay was calculated using the formula of cell death percentage. From the calculated of cell death percentage it was found that treatment 3 with concentration of 20 μg / ml could inhibit the highest cell growth. Then the percentage data of cell death was used to obtain IC50 value as 50% inhibition value to the treatment. In this research, IC50 average of all treatments is 13,496 μg / ml. The combined 5-fluorouracil and coelomic fluid cytotoxic tests of Lumbricus rubellus against HT-29 cell line were examined using Kruskal-Wallis method. In Kruskal Wallis test, the data has significant value (p <0,05) so that the data is valid to be used as research data. Based on this research, it can be concluded that the combination of 5-fluorouracil and coelomic fluid from Lumbricus rubellus can increase the cytotoxic effect on cell line HT-29 with concentration of coelomic fluid 20 μg / ml has the highest cytotoxic effect compared to all treatments and IC50 value from all treatment a combination of 5-fluorouracil and coelomic fluid from Lumbricus rubellus 13.496 μg / ml.
