Abstract
Saat ini sebagian besar makanan yang dikonsumsi selain mengandung bahan penyedap seperti MSG juga mengandung lemak yang tinggi. Banyak penelitian pada hewan coba yang menunjukkan bahwa mengonsumsi MSG yang berlebihan dapat memicu beberapa penyakit kronik seperti diabetes mellitus tipe 2, penyakit kardiovaskular, efek neurotoksis pada otak, dan gangguan sistem reproduksi. Selain MSG, diet tinggi lemak juga memiliki dampak negatif pada tubuh. Penyimpanan lipid yang berlebihan dalam tubuh dapat menginduksi gangguan fungsi ovarium. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian kombinasi diet tinggi lemak dan MSG terhadap jumlah folikel de Graaf dan atretik. Studi eksperimental menggunakan metode rancangan Randomized control group post test design, dilakukan pada tikus Wistar betina. Sampel dipilih secara acak dan dibagi dalam 6 kelompok, yaitu kontrol negatif (-), kontrol (+) 1 diet tinggi lemak (DTL), kontrol (+) 2 MSG 0,7mg/gBB, Perlakuan 1 (DTL+MSG 0,05mg/gBB), Perlakuan 2 (DTL+MSG 0,2mg/gBB), dan Perlakuan 3 (DTL+MSG 0,35mg/gBB). Masing-masing kelompok terdiri dari 5 ekor tikus. Penelitian dilakukan selama 56-61 hari. Variabel yang dihitung adalah jumlah folikel de Graaf dan Atretik dengan menggunakan mikroskop cahaya, perbesaran 400x. Hasil penelitian dengan menggunakan uji ANOVA menunjukkan tidak ada perbedaan yang bermakna pada penurunan jumlah folikel de Graaf dalam kelompok (p=0,06), sedangkan terdapat perbedaan yang bermakna pada peningkatan jumlah folikel atretik masing-masing kelompok (p=0,001). Kesimpulan dari penelitian ini adalah pemberian kombinasi diet tinggi lemak dan MSG berbagai dosis menimbulkan efek yang sama dengan efek yang ditimbulkan oleh diet tinggi lemak saja atau MSG dosis tinggi terhadap penurunan jumlah folikel de Graaf dan peningkatan folikel atretik.
English Abstract
Currently, most foods consumed, in addition to containing food seasonings such as MSG also contains high fat. Many experimental animal studies have shown that excessive consumption of MSG can trigger some chronic diseases such as type 2 diabetes mellitus, cardiovascular disease, neurotoxic effects on the brain, and reproductive system disorders. In addition to MSG, a high-fat diet also has a negative impact on the body. Excessive lipid storage in the body may induce ovarian function impairment. This study aims to determine the effect of combining high-fat diet and MSG on the number of de Graaf and atretic follicles. The experimental study used the Randomized control group post test design, performed on female Wistar rats. The sample was randomly selected and divided into 6 groups, consists of negative control (-), control (+) 1 high-fat diet (HFD), control (+) 2 MSG 0.7mg / gBB, Treatment 1 (HFD + MSG 0.05mg / gBB), Treatment 2 (HFD + MSG 0.2mg / gBB), and Treatment 3 (HFD + MSG 0.35mg / gBB). Each group consists of 5 rats. The study was conducted for 56-61 days. The calculated variable are number of de Graaf and atretik follicles by using a light microscope, 400x magnification. Results of the study using ANOVA test showed no significant difference in decreasing number of de Graaf follicles in the group (p = 0.06), whereas there was a significant difference in increasing the number of atretic follicles of each group (p = 0.001). Conclusions of this study were the combination of a high-fat diet and various doses of MSGs having the same effect as the effects of high-fat diet alone or high-dose MSG on decreased follicular counts of de Graaf and increased atretic follicles.
