Abstract

Latar Belakang: Fibrosis hati merupakan hasil proses fibrogenesis dan fibrolisis. Pada proses fibrogenesis, HSCs memproduksi MMP-2 dan TIMP-2. Keduanya berfungsi mengatur homeostasis matriks ekstraseluler. Kurkumin meningkatkan ekspresi MMP-2, menurunkan ekspresi TIMP-2 serta mendorong apoptosis HSCs, yang dengannya fibrogenesis menjadi terhambat. Peran kurkumin, ekspresi MMP-2 dan ekspresi TIMP-2 terhadap HSCs dalam proses fibrolisis belum banyak diteliti. Tujuan: Mengetahui hubungan pemberian kurkumin terhadap peningkatan ekspresi MMP-2 dan penurunan ekspresi TIMP-2 di HSCs tikus model fibrosis hati. Metode: Penelitian ini menerapkan metode eksperimental pada tikus jantan strain wistar. Terdapat 8 kelompok tikus yakni KN, KP2, KP5, KP9, dan KK2, KK5, KK9. Masing-masing terdiri dari 4 tikus sampel. KP dipapar CCl4 1cc/kgbb 2x perminggu intraperitoneal selama 9 minggu membentuk fibrosis F3. KN diinjeksi Normal Salin. Setelah diinjeksi CCl4, Kelompok Kontrol (KK2,KK5,KK9) diberikan plasebo dan Kelompok Perlakuan (KP2,KP5,KP9) diberikan kurkumin. Analisis ekspresi MMP-2 dan ekspresi TIMP-2 menggunakan uji t independen, ANOVA dan korelasi. Hasil: Terdapat peningkatan ekspresi MMP-2 pada KK9 dan penurunan ekspresi TIMP- 2 pada KK5 dan KK9; peningkatan ekspresi MMP-2 dan penurunan ekspresi TIMP-2 pada KP2, KP5, dan KP9. Kesimpulan: Peningkatan signifikan ekspresi MMP-2 dan penurunan signifikan ekspresi TIMP-2 pada tikus yang diberikan kurkumin selama 2 minggu, 5 minggu dan 9 minggu. Lama pemberian kurkumin berkorelasi dengan peningkatan ekspresi MMP-2 dan penurunan ekspresi TIMP-2 pada tikus model fibrosis hati

English Abstract

Background: Liver fibrosis result of fibrogenesis and fibrolysis. In fibrogenesis, MMP-2 and TIMP-2 produced by HSCs, regulate ECM homeostastic. Curcumin inhibit MMP-2, TIMP-2 expression and enhance HSCs apoptosis, which is inhibit fibrogenesis. Role of curcumin, MMP- 2 expression, and TIMP-2 expression in a fibrolysis has not been widely studied. Objective: Determine correlation between giving of curcumin to increase MMP-2 expression and decreased TIMP-2 expression rat model of liver fibrosis. Methods: Experimental study in male rats wistar strain. There are 8 groups KN, KP2, KP5, KP9, dan KK2, KK5, KK9; 4 rats for each. KP group was exposed to CCl4 1cc/kg intraperitoneally 2 times per week for 9 weeks to form F3 fibrosis. KN group was injected Normal saline. After CCl4 injected, Control group (KK) was given placebo and intervention group (KP) was given curcumin. Analysis of MMP-2 and TIMP-2 expression at HSCs is conducted by independent t-test, ANOVA and correlation test. Result: Increase MMP-2 expression in KK9 and decrease TIMP-2 expression in KK5 and KK9. Increase MMP-2 expression and decrease TIMP-2 expression in KP2, KP5, and KP9. Conclusion: Remarkable increase MMP-2 and remarkable decrease TIMP-2 expression in HSCs after giving of curcumin 2, 5 and 9 weeks. Duration curcumin correlated with increase MMP-2 expression and decrease TIMP-2 expression in rat model liver fibrosis
