Abstract

Sektor pertanian saat ini mulai krisis akibat pertambahan penduduk yang mengakibatkan berkurangnya vegetasi dan pasokan serasah pada permukaan tanah. Efek dari alih fungsi lahan dengan pengolahan lahan yang intensif yang akan merubah menjadi lahan marginal sehingga mempengaruhi kesuburan tanah. Kesuburan tanah berkaitan erat dengan biota di dalam tanah terutama bakteri dan tersedianya unsur hara bagi tanaman. Salah satu bakteri yang mampu mencukupi kebutuhan unsur hara esensial berupa N untuk menjaga kesuburan tanah adalah bakteri penambat nitrogen nonsimbiotik. Tujuan dari penelitian yaitu mengetahui pengaruh penggunaan lahan terhadap diversitas dan populasi bekteri penambat nitrogen nonsimbiotik di berbagai penggunaan lahan dan mengetahui faktor lingkungan yang berpengaruh terhadap diversitas dan populasi bakteri penambat nirogen. Penelitian dimulai pada bulan Oktober 2017 - April 2018. Penelitian dilakukan dengan metode Survei pada lokasi penelitian di UB Forest, teridiri dari dua lokasi yaitu Dusun Sumbersari, Desa Tawangargo dan Dusun Buntoro, Desa Bocek, Kecamatan Karangploso, Kabupaten Malang. Pengambilan sampel diambil pada 5 penggunaan lahan yaitu Kawasan lindung (KL), Mahoni kopi (MK), Mahoni + talas (MS), Pinus kopi (PK) dan Pinus + wortel (PS) dengan 3 kali pengulangan sehingga menghasilkan 15 kombinasi perlakuan. Pengambilan sampel dengan luas 20 meter x 20 meter. Parameter yang dimati terdiri dari biologi tanah (bakteri penambat nitrogen Azotobacter sp, populasi bakteri, uji patogenitas), lingkungan (suhu, kelembaban, berat dan ketebalan serasah) dan kimia tanah terdiri dari (kadar air, pH, C-organik, N-total, Amonium dan Nitrat tanah). Data diolah menggunakan GenStat dengan rancangan tersarang (Nested) uji korelasi dengan taraf 5%. Hasil penelitian menunjukkan diversitas dan populasi bakteri penambat nitrogen nonsimbiotik (Azotobacter sp) berpengaruh nyata pada berbagai penggunaan lahan di UB Forest. Populasi bakteri tertinggi pada kawasan lindung (KL) dengan jumlah rerata 111,92 x 107 cfu/g dan penggunaan lahan mahoni kopi memiliki jumlah bakteri terendah dengan rerata 37,5 x 107 cfu/g. Faktor lingkungan seperti kerapatan tajuk, berat dan ketebalan serasah mempengaruhi diversitas dan populasi bakteri penambat nitrogen nonsimbiotik di UB Forest. Karakteristik sifat kimia seperti C-organik dan N-total turut mendukung diversitas dan populasi bakteri penambat nitrogen nonsimbiotik. Isolat yang ditemukan sebanyak 29 dari berbagai penggunaan lahan, terdiri dari kawasan lindung (KL) 8 isolat, mahoni + kopi (MK) 4 isolat, mahoni + talas (MS) 6 isolat, pinus + kopi (PK) 6 isolat dan pinus + wortel (PS) 4 isolat. Hasil uji patogenitas negatif untuk ke 8 isolat yang terkarakterisasi.

English Abstract

Increasing population growth can affect of crisis agricultural sector included decreasing vegetation and soil surface litter supply. The effects of landuse with intensive land management can change land becomes marginal and affecting soil fertility. Soil fertility is related to biota in the soil, particularly bactery and available soil nutrients for plant. One of the bactery can suffice the need of essential nutrients such as Nitrogen to maintain soil fertility is nonsimbiotic nitrogen-fixing bacteria. The aims of this research is to know the effect of landuse diversity and nonsimbiotic nitrogen-fixing bacteria population in various landuses, and to know the influences of environment factor to diversity and nonsimbiotic nitrogen-fixing bacteria population. The research was conducted in October 2017 - April 2018. This research used survey method at UB Forest, consist of two locations, Dusun Sumbersari, Tawangargo Village and Dusun Buntoro, Bocek Village, Karangploso Subdistrict, Malang Regency. Sampling was taken on 5 land uses i.e. Protected Area (KL), Mahogany + Coffee (MK), Mahoni + Taro (MS), Pinus + Coffee (PK) and Pinus + Carrot (PS) with 3 repetitions resulting in 15 combinations of treatments. Area of sampling of 20 meters x 20 meters. The observed parameters of soil biology (Azotobacter sp nitrogen-fixing bacteria, bacterial population, pathogenicity test), environment factor (temperature, humidity, weight and thickness of litter) and soil chemistry consist of (moisture content, pH, C-organic, N-total, Ammonium and soil Nitrate). The data were processed using GenStat edition 17th with Nested of correlation test with 5% level. The results showed that diversity and population of nonsimbiotic nitrogenfixing bacteria (Azotobacter sp) had significant effect on various landuses. The highest bacterial populations was found in protected areas (KL) with the average number of 111,92 x 107 cfu/g x 107 and mahogany + coffee had the lowest bacterial with the average number of 37.5 x 107 cfu/g x 107. Environmental factors such as crown density, weight and litter thickness affect the diversity and population of nonsimbiotic nitrogen fixing bacteria in UB Forest. Characteristics of chemical properties such as C-organic and N-total also support the diversity and population of nonsimbiotic nitrogen fixing bacteria. The isolates found were 29 of the various land uses, consisting of 8 isolates, mahogany + coffee (MK) 4 isolates, mahogany + talas (MS) 6 isolates, pine + coffee (PK) 6 isolates and pine + carrots (PS) 4 isolates. The result of negative pathogenic test for the 8 isolates characterized.
