Abstract

Hiperlipidemia merupakan peningkatan konsentrasi lipid (trigliserida, kolesterol ataupun keduanya). Pemberian pakan tinggi lemak merupakan salah satu penyebab terjadinya hiperlipidemia. Kondisi tersebut menyebabkan meningkatnya kadar LDL, kolesterol total serta kerusakan organ aorta. Pengendalian hiperlipidemia dapat dilakukan dengan konsumsi antioksidan. Ekstrak Air biji petai cina (Leucaena leucocephala) mengandung flavonoid yang dapat digunakan sebagai antioksidan. Penelitian ini bertujuan untuk mengetahui pengaruh terapi ekstrak air biji petai cina(Leucaena leucocephala) terhadap kadar kolesterol total dan histopatologi aorta hewan model tikus (Rattus norvegicus) hiperlipidemia. Penelitian ini menggunakan 20 ekor tikus jantan, strain Wistar umur 10-12 minggu dengan berat badan 110-130 gram. Pembuatan keadaan hiperlipidemia dilakukan dengan cara pemberian diet Propiltiourasil (PTU) 0,02 % dan kuning telur 10 ml/kg BB yang diberikan secara force feeding selama 10 hari, sedangkan pemberian terapi ekstrak air biji petai cina dengan dosis 0,25 g/kg BB, 0,5 g/kg BB dan 1 g/kg BB diberikan secara oral selama 14 hari. Pengukuran kadar kolesterol total menggunakan metode CHOD-PAP (Enzymatic-spechtophotometric) dan pembuatan preparat histopatologi aorta menggunakan metode pewarnaan Hematoksilin Eosin (HE). Analisis data kuantitatif menggunakan one way Analisis of Variance (ANOVA) dengan uji lanjutan Tukey (α = 5 %). Hasil penelitian menunjukkan bahwa terapi ekstrak air biji petai cina mampu menurunkan kadar kolesterol total secara signifikan (p<0,05). Dosis 1 g/kg BB merupakan dosis terbaik dalam menurunkan kadar kolesterol total serta dapat memperbaiki kerusakan aorta tikus model hiperlipidemia. Kesimpulan dari penelitian ini adalah ekstrak air biji petai cina dapat menurunkan kadar kolesterol total dan memperbaiki kerusakan aorta pada hewan model tikus hiperlipidemia.

English Abstract

Hyperlipidemia was an increasing concentration of lipids (triglycerides, cholesterol, or both). Fatty food and propiltiourasil (PTU) caused hyperlipidemia which followed by the increasing of LDL, total cholesterol level and aorta histopatology. Hyperlipidemia animal model was done done by consumption of antioxidants. Water extract of the leucaena’s seeds (Leucaena leucocephala) contains flavonoids used as antioxidants. The aim of this research was to know the effect of leucaena’s seed (Leucaena leucocephala) water extract based on decreasing total cholesterol levels and aorta histopathology changes confirmed by reducing of fat accumulation on rat (Rattus norvegicus). This study used 20 rats, Wistar strain aged of 10-12 weeks with 110-130 grams of body weight. Hyperlipidemia animal model was done by giving Propylthiouracil (PTU) 0.02% and egg yolks 10 mL / kg BW administered by force feeding for 10 days, while the water extract of Leucaena’s seed therapy dose 0.25 g/kg BW, 0.5 g/kg BW and 1 g/kg BW given orally for 14 days. The parameters observed in this study was Total Cholesterol levels and aorta histopathology. Measurement of Total Cholesterol was calculated by using CHOD-PAP methods (Enzymaticspechtophotometric) and aorta histopathology were stained using Hematooksilin Eosin methods (HE). The analysis of quantitative data using one way Analysis of Variance (ANOVA) followed by Tukey test (α = 5%). The results showed that water extract of leucaena’s seed therapy could reduce total cholesterol significantly (p<0,05). Dose of 1 g/kg BW was the better dose of this research that reduced total cholesterol levels. Water extract of Leucaena’s seed had repair aorta histopathology. Conclusion this study was water extract of leucaena’s seed therapy could decrease the total cholesterol levels and repair aorta histophatology of hyperlipidemia rats
