Abstract

Perempuan usia 6-18 tahun Di Sanga-Sanga Muara Kutai Kartanegara Kalimantan Timur mengeluh dismenorhea primer. Masyarakat sekitar termasuk remaja konsumsi mata air dan aliran Sungai Mahakam yang mengandung kadmium 0.06 mg/L. Toksisitas kadmium menjadi sinyal kaskade yang mengganggu stabilitas membrane lisosom dan meningkatkan pelepasan enzim fosfolipase-A2 induce sintesis prostaglandin serta toksisitas melalui transport ”mimicry kalsium ion” dimana ion kalsium merupakan unsur regulator untuk proses seluler termasuk kontraksi otot, mediator kerja hormon dan aktifitas enzim serta mengganggu steroidogenesis. Tujuan penelitian ini untuk membuktikan dampak pemberian kadmium terhadap penurunan kadar progesteron dan peningkatan ekspresi PGF2∝ jaringan uterus tikus betina galur wistar. Penelitian eksperimental laboratorik post test only with control group pada tikus betina galur wistar. Tikus dibagi dalam kelompok kontrol negatif dan kelompok perlakuan pemberian Cadmium Acetate Dihydrate personde konsentrasi 0.03; 0.06; 0.09 ppM setelah 42 hari di swab vagina dan dibedah pada fase diestrus selanjutnya dilakukan pemeriksaan kadar progesteron serum elisa kit sedangkan ekspresi PGF2∝ uterus dengan immunohistokimia anti PGF2∝. Analisa data dengan uji Shapiro-Wilk, uji Anova One Way, uji regresi linier sederhana dan korelasi Pearson. Hasil penelitian menunjukkan bahwa pemberian Cadmium Acetate Dihydrate konsentrasi 0.03; 0.06; 0.09 ppM menurunkan kadar progesteron nilai p=0.000 0.05 namun uji LSD menunjukkan penurunan kadar progesteron serum tidak tergantung konsentrasi, Peningkatan ekspresi (PGF2∝) zona basal nilai p=0.000 0.05, stratum vascular nilai p=0.000 0.05. Pengaruh penurunan kadar progesteron serum dipengaruhi peningkatan ekspresi PGF2∝ di zone basal sebesar 0.360 dan stratum vascular sebesar 0.555. Penelitian ini membuktikan pemberian Cadmium Acetate Dihydrate dapat menurunkan kadar progesteron dan meningkatkan ekspresi PGF2∝ jaringan uterus tikus betina galur wistar pada bagian endometrium (Zona Basal) dan miometrium (Stratum Vasculer).

English Abstract

Women aged 6-18 years in Sanga-Sanga Muara Kutai Kartanegara in East Kalimantan to complains primary dysmenorhea. Communities around including teenagers consumption of of springs and Mahakam River which contains cadmium 0.06 mg / L. Cadmium toxicity also function as signalling cascade that disturbs lisosome membrane stability and increase release of fosfolipase-A2 enzyme which promote syn sis of prostaglandin. A heavy metal, cadmium may cause organ and reproductive system toxicity by transport mechanism of “calsium ionic mimicry” in which ionized calcium serves as regulator of cellular processes such as muscle contraction, coagulation, enzyme activity, membrane irritability and mediator of hormone, as well as interfere with steroidogenesis. goal of this research is to prove effect of giving Cadmium Acetate Dihydrate to decreasing of progesteron rate, increasing PGF2∝ expression of uterine tissue of female rats wistar strain. This is experimental laboratory post-test only control group using Rattus norvegicus of wistar strain female rats. rats were divided into negative control group and treatment group which got 0.03; 0.06 and 0.09 ppM of Cadmium Acetate Dihydrate. Cadmium Acetate dyhidrate was administered orally using sonde for 42 days. After treatment, rats were subject to vaginal swab and surgery at diestrus phase. Next, Elisa kit was used to determine level of serum progesteron, while immunohitochemical localization of anti PGF2∝ was used to determine expression of prostaglandin F2α (PGF2∝) of rats`uterus. data were analyzed using Shapiro-Wilk, One Way Anova (uji F), simple linear regression analysis, and Pearson correlation test. finding of research indicated that administration of Cadmium Acetate Dihydrate of concentration 0.03 ppM; 0.06 ppM; 0.09 ppM for 42 days could decrease progesteron level (p=0.000 0.05). However, data were analyzed using LSD decrease in level of serum progesteron did not depend on concentration of Cadmium Acetate Dihydrate, on increase of expression (PGF2∝) of basal zone (p=0.000 0.05), stratum vascular and (p=0.000 0.05). decrease of serum progesteron level was due to increase of PGF2∝ expression in basal zone of 0.360 dan stratum vascular of 0.555. This research was proving administration of Cadmium Acetate Dihydrate in female rats of wistar strain decreases serum progesteron level, although decrease does not depend on concentration of Cadmium Acetate Dihydrate. administration of Cadmium Acetate Dihydrate in female rats of wistar strain increases expression of PGF2α in rat`s utery of endometrial tissue (Basal Zone) and miometrium (Stratum Vasculer).
