Abstract
Komoditas perikanan air tawar yang berpeluang besar untuk ditingkatkan produksinya adalah ikan mas ( Cyprinus carpio ). Intensifikasi budidaya justru menimbulkan masalah baru dengan mewabahnya penyakit ikan yang disebabkan bakteri Aeromonas hydrophila. Pemberian imunostimulan dari senyawa bioaktif teripang ( Holothuria scabra) diduga dapat meningkatkan respon imun nonspesifik pada ikan mas sebagai mekanisme pertahanan diri terhadap serangan penyakit. Tujuan penelitian ini mengetahui peranan pemberian total fenol teripang pasir terhadap respon imun non spesifik ikan mas serta dosis optimal pemberian total fenol teripang pasir dalam meningkatkan respon imun non spesifik. Ekstraksi komponen bioaktif teripang dilakukan menggunakan metanol dan difraksinasi dengan pelarut etil asetat ( v/v ). Identifikasi total fenol pada fraksi etil asetat dilakukan menggunakan spektrofotometer UV-vis dan infrared. Hasil penelitian menunjukkan bahwa berdasarkan spektra UV-vis ekstrak teripang pasir merupakan senyawa fenolik dengan serapan kuat pada panjang gelombang 252 nm. Hasil spektra Infrared menunjukkan adanya fenol karena mengandung gugus hidroksil (OH) dan uluran C-O. Total fenol yang dihasilkan diujikan pada ikan mas dengan penyuntikan secara intraperitonial, konsentrasi 0; 0,09; 0,18; dan 0,27 mg fenol/kg ikan. Uji tantang dilakukan menggunakan bakteri Aeromonas hydrophila (10 7 sel/ml) melalui metode perendaman. Pengamatan parameter meliputi pengamatan hematologi dan total protein plasma. Data yang diperoleh dianalisis dengan menggunakan analisa keragaman satu arah ( one way anova ) menggunakan Minitab Statistical Software versi 14 untuk mengetahui perbedaan antar perlakuan. Analisa regresi dilakukan untuk mengetahui hubungan pemberian dosis perlakuan terhadap parameter yang diamati. Hasil pengamatan parameter hematologi ikan mas menunjukkan nilai hematokrit, hemoglobin, eritrosit, limfosit pra infeksi mengalami peningkatan dan post infeksi bakteri mengalami penurunan signifikan (p 0.05). Leukosit dan neutrofil pra dan post infeksi mengalami peningkatan signifikan (p 0.05). Eosinofil, monosit, kadar protein plasma pra dan post infeksi bakteri tidak berbeda nyata antar perlakuan (p 0.05). Pemberian total fenol dosis 0,09 mg/kg merupakan dosis optimal dalam meningkatkan respon imun non spesifik pada ikan mas (Cyprinus carpio ).
English Abstract
Freshwater fishery commodities are most likely to be increased production of carp ( Cyprinus carpio ). Intensification of cultivation which raises new problems with fish disease outbreaks caused by Aeromonas hydrophila . Providing of immunostimulatory bioactive compounds of sea cucumbers (Holothuria scabra) can enhance non-specific immune responses in the goldfish as a mechanism of defense against disease. The purpose of this study is to determine the role administration of total phenols sea cucumber with non specific immune parameters in carp and optimal dose of total phenols sea cucumber ( Holothuria scabra ) in enhancing nonspecific immune response. Extraction of bioactive components from sea cucumber prepared by using methanol and fractionated with ethyl acetate solvent (v/v). Identification of total phenols in ethyl acetate fraction performed using UV-Vis spectrophotometer and infrared. The result showed that according to UV-vis spectrometry of total phenols sea cucumber is the phenolic compound have strong absorption of light in wavelength 252 nm. Characterization of compounds via infrared spectroscopy identified that phenolic compound contain of hydroxyl group (OH) and C-O groups. Total phenol produced was tested on carp with intraperitonial injection, the concentration of 0.09, 0.18 and 0.27 mg phenol/kg fish. Challenge test was done by using the Aeromonas hydrophila (10 7 cells/ml) through the immersion method. The observations included hematological parameters and total plasma proteins. The data were expressed as arithmatic mean ± standard error (SE). Statistical analysis involved one-way analysis of variance (ANOVA) by MINITAB 14. The levels of significance were expressed as P-value less or greater than 0,05. The observations of hematological parameters that hematocrit, hemoglobin, erythrocytes, lymphocyte pre infection was increased significantly and post-bacterial infection was decreased significantly (p 0.05). Leukocytes, neutrophils pre-and post-infection were significantly increased (p 0.05). Eosinophils, monocytes, total protein pre-and post-bacterial infection were not differ significantly between treatment (p 0.05). The use of phenolic compounds at 0.09 mg/kg can be increase of non-specific immune response in carp ( Cyprinus carpio ).
