Abstract

Mikroalga Chlorella pyrenoidosa merupakan jenis mikroorganisme fotosintetik, terdapat komponen penyusun kandungan esensial (protein, lipid, klorofil dan karotenoid) yang dimanfaatkan oleh manusia dibeberapa sektor yaitu; sektor perikanan, indrustri makanan, farmasi, dan industri kosmetik. Tujuan dari penelitian ini untuk mengetahui pengaruh air kelapa sebagai substitusi pupuk pertumbuhan sel C. pyrenoidosa, kadar protein, lipid, klorofil, karotenoid, ukuran sel dan parameter kualitas air (suhu, pH, salinitas, nitrat dan fospat) di dalam medium kultur. Telah dilakukan Oktober 2013 sampai April 2014, untuk kultur dilakukan di Laboratorium Nutrisi, Fakultas Perikanan dan Ilmu Kelautan, pengukuran kadar protein, lipid, klorofil, karotenoid dan ukuran sel dilakukan dibeberapa Laboratorium Universitas Brawijaya Malang. Penelitian bersifat eksperimen dengan menggunakan rancangan acak lengkap (RAL) pola faktorial dengan 6 perlakuan, yaitu media menggunakan air kelapa dengan konsentrasi 0%, 3%, 5%, 7%, 9%, dan 11%. Hasil penelitian menunjukkan pertumbuhan sel tertinggi sebesar 8.375.000 Sel/ml, terjadi pada hari keenam dengan konsentrasi terbaik pada perlakuan 7%. Kadar protein tertinggi terdapat pada perlakuan konsentrasi air kelapa 7% dan terendah pada konsentrasi 9%. Kadar lipid tertinggi terjadi pada konsentrasi 3% dan terendah pada konsentrasi 11%. Kadar klorofil total dan karotenoid tertinggi terdapat pada konsentrasi 7% dan terendah pada konsentrasi 9%. Ukuran sel C. pyrenoidosa., memiliki ukuran terbesar yaitu 22,5 μm pada konsentrasi 7% dan yang terendah 12, 5 μm pada konsentrasi 11%, dan untuk paremeter kualitas air didalam media kultur yaitu : suhu berkisar antara 29 – 32 0C, pH berkisar antara 6,0 – 8,0. Salinitas berkisar antara 30 -35 ppm, dan untuk nitrat dan fosfat yang tertinggi pada konsentrasi 0%, terendah pada Konsentarsi 9% dan 11%.

English Abstract

Microalgae Chlorella pyrenoidosa is a type of photosynthetic microorganisms, are essential components of content (proteins, lipids, chlorophyll and carotenoids) that is used by humans in some sectors, namely; fisheries sector, indrustri food, pharmaceutical, and cosmetic industries. The purpose of this study was to determine the effect of coconut water as a fertilizer substitute C. pyrenoidosa cell growth, protein content, lipid, chlorophyll, carotenoids, cell size and water quality parameters (temperature, pH, salinity, nitrate and phosphate) in the culture medium. Has been done in October 2013 until April 2014, to culture performed at the Laboratory of Nutrition, Faculty of Fisheries and Marine Science, measuring levels of proteins, lipids, chlorophyll, carotenoids and cell size is done in several laboratories UB. Experimental study using a completely randomized design with 6 treatments factorial, ie media using coconut water with a concentration of 0%, 3%, 5%, 7%, 9%, and 11%. The results showed the highest cell growth amounted to 8.375 million cells/ml, occurred on the sixth day with the best concentration of 7% on treatment. Highest protein content found in coconut water treatment concentration of 7% and the lowest concentration of 9%. The highest lipid levels occurred at a concentration of 3% and the lowest at 11% concentration. Levels of total chlorophyll and carotenoid found in highest concentrations of 7% and the lowest concentration of 9%. Cell size of C. pyrenoidosa., Has the largest size is 22.5 lm at a concentration of 7% and the lowest was 12, 5 lm at a concentration of 11%, and for water quality parameter in culture medium, namely: temperature ranges between 29-32 0C, pH ranged from 6.0 to 8.0. Salinity ranges between 30 -35 ppm, and for the highest nitrate and phosphate at a concentration of 0%, the lowest concentration of 9% and 11%.
