Abstract

Tujuan dari penelitian ini adalah mengetahui efek alfa-pinen terhadap apoptosis neuron pada kultur campuran neuron-glia otak tikus setelah pemberian protein HIV-1 gp120. Prosedur penelitian ini adalah isolasi dan kultur neuron-glia otak tikus, setelah sel tumbuh merata, diberikan gp120 dan/atau alfa-pinen (Kontrol; gp120 1 nM; alfa-pinen 10 µg/mL; alfa-pinen 100 µg/mL; alfa-pinen 10 µg/mL + gp120 1 nM; alfa-pinen 100 µg/mL + gp120 1 nM, dan alfa-pinen 200 µg/mL + gp120 1 nM. Gp120 diberikan selama 1 jam dan alfa-pinen diberikan selama 2 jam. Setelah itu dilakukan deteksi apoptosis menggunakan Terminal deoxynucleotidyl transferase-mediated biotinylated UTP nick end labeling (TUNEL) dan imunositokimia. Sel yang telah terwarnai dihitung dan dianalisis dengan ANOVA, uji lanjut Duncan dengan program SPSS. Hasil penelitian menunjukkan bahwa pemberian alfa-pinen 10 µg/mL dan 100 µg/mL menunjukkan terjadinya apoptosis neuron berturut-turut 34,87% dan 36,59%. Pemberian 10 µg/mL alfa-pinen + 1 nM gp120 menimbulkan apoptosis neuron sebesar 25,45%. Pemberian alfa-pinen 100 µg/mL + gp120 1 nM terjadi apoptosis sejumlah 38,74%. Pemberian alfa-pinen 200 µg/mL + 1 nM gp120 menunjukkan adanya apoptosis neuron sebanyak 28,21%. Penelitian ini menunjukkan bahwa alfa-pinen dapat menyebabkan apoptosis neuron pada kultur neuron glia tanpa atau dengan paparan gp120. Pada kultur neuron-glia yang tidak dipapar gp120, alfa-pinen tergantung dosis menyebabkan apoptosis. Semakin tinggi dosis alfa-pinen, jumlah neuron yang mengalami apoptosis semakin meningkat. Pada kultur neuron-glia yang telah dipapar gp120, alfa-pinen dosis 10 µg/mL menimbulkan jumlah apoptosis neuron lebih sedikit dibandingkan alfa-pinen dengan dosis 100 µg/mL maupun 200 µg/mL. Secara keseluruhan dapat disimpulkan bahwa pemberian alfa-pinen pada sistem neuron-glia normal dapat meningkatkan apoptosis, sedangkan apabila sistem neuron-glia dalam keadaan terpapar gp120, alfa-pinen dosis kecil dapat menurunkan apoptosis neuron.

English Abstract

Apoptosis or cell death, is important physiological process for normal cellular replacement. Apoptosis is celluler respon to infection such as virus infection, Human Immunodeficiency Virus (HIV), adenovirus, Epstein-Barr virus and baculovirus. The aim of this research was to study the effect of alpha-pinene to neuron apoptosis in mixed neuron-glia cells culture of rat brain gp120 protein exposure. This research procedure, neuron cells were isolated from mix neuron-glia cells culture of rat brain. Cells was exposured with alpha-pinene at 1 hour and gp120 at 2 hours. The groups of cells culture were devided one control group and 6 treatments group: 1nm of gp120 protein, 10 µg/mL alpha-pinene, 100 µg/mL alpha-pinene, 200 µg/mL alpha-pinene, 10 µg/mL alpha-pinene+1nm of gp120 protein, 100 µg/mL alpha-pinene+1nm of gp120 protein and 200 µg/mL alpha-pinene+1nm of gp120 protein. Apoptesis cells was identified by using terminal deoxynucleotidyl transferase-mediated biotinylated-UTP-nick-end labeling (TUNEL) and immunocytochemistry with MAP-2 rabbit polyclonal lgG antiserum. This research result is showed that alpha-pinene is increasing neuron cells apoptosis in neuron-glia cells culture with or without gp120. Alpha-pinene with doses 10 µg/mL and 100 µg/mL are increased cells apoptosis. However cells apoptosis in neuron cells induced by 1nM of gp120 protein + 200 µg/mL of alpha-pinene was decreased than other treatments. This research indicated that increasing neuron cells apoptosis induced by alpha-pinene.
