Abstract

Studi mengenai pemanfaatan katalis biotik lipase dari Rhizomucor miehei untuk mengkatalisis reaksi siklisasi-asilasi sitronelal telah dilakukan. Pada penelitian ini dilakukan reaksi siklisasi-asilasi sitronelal menggunakan sumber asil asetat anhidrida dan asetat glasial dalam pelarut n-heksan pada kondisi temperatur kamar dan temperatur 50oC pada berbagai variasi waktu. Hasil yang diperoleh yaitu reaksi tidak dapat menghasilkan produk isopulegil asetat sebagai produk utama. Enzim lipase hanya mampu mengkatalisis reaksi siklisasi sitronelal menjadi isopulegol. Konversi sitronelal mencapai 20,64% pada temperatur ruang dengan lama reaksi 24 jam dan pada temperatur 50 oC mencapai 9,51% dengan lama reaksi delapan jam. Nilai selektivitas lipase masing-masing dengan lama reaksi 12 jam mencapai 73,13% pada temperatur ruang dan mencapai 34,10% pada temperatur 50 oC. Konversi sitronelal mengalami peningkatan dengan bertambahnya waktu namun selektivitas pembentukan isopulegol menurun. Selektivitas pembentukan isopulegol yang rendah tidak cukup menyediakan substrat bagi enzim untuk mengkatalisis reaksi asilasinya menjadi isopulegil asetat.

English Abstract

Rhizomuchor miehei lipase enzyme-catalyzed reaction of Citronellal has been done. Two consecutive reactions of cyclization and acylation on Citronellal was performed by using acetic anhydrous acid and acetate glacial as acyl agent, Rhizomuchor miehei lipase enzyme and n-hexan as a solvent. Reactions were conducted at room temperature and 50 oC. The result was shown that isopulegyl acetate as the final product could not be produced. It can be happened because the value of isopulegol as cyclization product was not quite enough to play role as a substrat for acylation reaction. The enzyme only catalyze the cyclization reaction. The best convertion of substrat and selectivity to product, isopulegol, at room temperature were 20,64% (24 hrs) and 73,13% (12 hrs). On the other hand at 50 oC, convertion and selectivity were 9,51% (8 hrs) and 34,10% (12 hrs). The convertion and selectivity were very affected by time of reactions. It can be concluded that the longer time of reactions would increase the value of convertion, but the value of selectivity was decrease.
