Abstract

Porang (A. muelleri Blume) merupakan anggota famili Araceae yang mengandung kristal kalsium oksalat (CaOx) dan memiliki beberapa periode tumbuh. Periode tumbuh berbeda diduga berpengaruh terhadap kerapatan kristal CaOx. Penelitian ini bertujuan untuk mengetahui kerapatan sel dan kristal CaOx total serta jenis kristal pada umbi hasil periode tumbuh berbeda. Sampel umbi dari tiga periode tumbuh yang berbeda diperoleh dari Madiun. Sampel umbi ditimbang kemudian dicuplik menggunakan ‘corkbore’ dari arah tepi menuju tengah. Pada sisi tepi dan tengah masing-masing dibuat tiga irisan tipis untuk preparat. Irisan umbi dijernihkan menurut metode Ilarsan yang dimodifikasi. Pengamatan morfologi kristal dan penghitungan kristal menggunakan mikroskop cahaya (perbesaran 100x). Data yang diperoleh dianalisis anova untuk mengetahui pengaruh periode tumbuh, bagian umbi, dan interaksi anatra periode tumbuh dan bagian umbi dengan menggunakan SPSS 16 for windows. Dengan menggunakan program yang sama dilakukan analisis regresi korelasi antara berat umbiterhadap kerapatan sel atau kerapatan kristal CaOx total serta analisis korelasi bivariate antara kerapatan sel terhadap kerapatan kristal. Hasil yang diperoleh menunjukkan bahwa periode tumbuh berbeda tidak berpengaruh terhadap kerapatan sel, kristal CaOx total serta jenis kristal CaOx (rafida dan druse). Bagian umbi berbeda berpengaruh signifikan terhadap kerapatan sel dan kristal CaOx total serta jenis kristal CaOx (rafida dan druse). Terdapat pengaruh interaksi antara periode tumbuh dan bagian umbi terhadap kerapatan sel dan kristal CaOx. Berat umbi tidak berkorelasi terhadap kerapatan sel maupun kristal CaOx total, namun terdapat korelasi antara kerapatan sel dan kristal CaOx total dengan nilai R = -0,597.

English Abstract

Porang (A. muelleri Blume) is group of families of Araceae with some growth period that contains (CaOx) crystals. The different growth period could influence to density of CaOx crystals. The aim of the research was to know density of cell, density of CaOx crystals and various form of CaOx crystals from different growth period. The tuber which have three growth periode were obtained from Madiun. The first step of the research weigh each tuber, then took the tissue from edge to center using cork bore. The very thin sliced tissue were provided by cutting both the edge and center direction using hand clam microtome. Each end direction was cut three pieces. Ilarsan modified method was applied to clear tissue. Observation crystal morphology and counting density of crystal were conducted of microscope (100x magnification). The obtained data were analyzed using SPSS 16 for windows. The analyzing include anova on density of cell, crystal CaOx, raphide or druse crystals, regression and correlation between tuber weigh and cell density or crystal density, and bivariate correlation between cell density and crystal density. The result showed that growth period was not significantly different to influence density of cell, CaOx CaOx crystal, raphide and druse crystals. The other hand, these fix variable were influenced significantly different under part of tuber (edge and center). Interaction part of tuber and growth periode only influenced significantly on raphide crystals . Regression and correlation result was not found between tuber weigh and cell density or CaOx crystal density. However bivariate correlation was found between cell density and CaOx crystal which R value was R= -0,597.
