Abstract

Ampas tahu mengandung protein sebesar 23,7 % dan mengandung bakteri proteolitik. Bakteri proteolitik dapat meningkatkan efisiensi penggunaan pakan ternak. Penelitian ini bertujuan untuk memperoleh isolat bakteri asal ampas tahu yang memiliki aktivitas proteolitik dan mempelajari aktivitas protease yang dihasilkan oleh bakteri proteolitik pada substrat bekatul. Penelitian ini menggunakan Rancangan Acak Kelompok (RAK) dengan dilakukan tiga kali pengulangan. Tahapan penelitian meliputi isolasi bakteri proteolitik dari ampas tahu, pengujian aktivitas bakteri proteolitik pada media calcium casseinate agar (CCA), karakterisasi bakteri, uji patogenisitas, pembuatan kurva pertumbuhan dan pengujian aktivitas proteolitik bakteri pada media bekatul 2 %. Data luas zona bening pada media CCA dianalisis menggunakan analisis ragam (Anova) dan dilanjutkan dengan uji Tukey, sedangkan aktivitas protease isolat bakteri pada media bekatul 2 % dianalisis menggunakan uji Independent Sample T Test (α = 0,05). Isolasi bakteri dari ampas tahu diperoleh 10 isolat. Isolat TP5K1 dan TP6K5 tidak patogen ditunjukkan dengan tidak adanya zona bening pada media blood agar, serta menunjukkan aktivitas protease tidak berbeda selama masa inkubasi pada media bekatul 2 % (p>0,05). Aktivitas protease tertinggi isolat TP5K1 yaitu sebesar 2,24 Unit/mL dengan jumlah sel bakteri 5,13 x 107 sel/mL, sedangkan aktivitas protease tertinggi isolat TP6K5 sebesar 2,37 Unit/mL dengan jumlah sel bakteri 5,26 x 107 sel/mL.

English Abstract

Tofu waste contains 23.7 % of protein and possible containing proteolytic bacteria. Proteolytic activity of bacteria could increase the efficiency of feed utilization and helps increase the absorption of amino acids by the livestock body.This research was carried out in order to get isolates bacteria in tofu waste having proteolytic activity and to study proteolitic activity of bacteria from tofu waste in rice bran substrate. Methods used in this research were, isolation of proteolytic bacteria from tofu waste, assay of bacterial protease activity on the calcium casseinate agar (CCA) medium, characterization of pathogenicity, preparation of bacterial growth curve and protease activity assay was performed in 2 % of rice bran medium. The wide of clear zone data of isolates bacteria were analyzed using Analysis of Variance (Anova) and continued by Tukey’s Test (α = 0.05), whereas protease activity of isolates bacteria on 2 % rice bran medium were analyzed using Independent Sample T-Test (α = 0.05). Isolation of bacteria from tofu waste resulted 10 isolates. Isolates TP5K1 and TP6K5 were not pathogen that shown by the absence of clear zone on blood agar medium. Protease activity isolates TP5K1 and TP6K5 were did not differ during the incubation period in the 2 % rice bran medium (p>0.05). The highest protease activity of isolate TP5K1 was 2.24 Unit/mL with the cell number of bacteria is 5.13 x 107 cell/mL, whereas the highest protease activity of isolate TP6K5 was 2.37 Unit/mL with the cell number was 5.26 x 107 cell/mL.
