Abstract

Kinerja biosensor didasarkan pada reaksi hidrolisis diazinon menjadi O,O dietil fosforotioat, 2-isopropil-6-metil pirimidin-4-ol dan ion H+ yang dikatalisis oleh alkalin fosfatase (ALP). Kinerja biosensor dipengaruhi oleh jumlah ALP teramobil, luas permukaan elektroda dan metode amobilisasi enzim. Pada penelitian ini alkalin fosfatase diamobilkan pada membran kitosan termodifikasi glutaraldehid yang terlapis di permukaan elektroda SPCE. Pengukuran diazinon dilakukan pada pH larutan 8,5 dengan jumlah ALP amobil 0,325; 0,650; 0,975; 1,30; 1,65 mg. Hasil penelitian menunjukan kinerja optimum diperoleh pada jumlah ALP amobil 1,65 mg secara ikatan silang, dengan kisaran konsentrasi (0,27 - 1,5) ppm, kepekaan 32,61 mV/ppm, batas deteksi 0,27 ppm dan dapat digunakan untuk mengukut 6 larutan sampel.

English Abstract

Biosensor’s performance is based on hydrolysis reaction of diazinon into O,O diethyl phosphorothioic , 2-isopropyl-6-methylpyrimidin-4-ol, and H+ catalyzed by alkaline phosphatase (ALP). Biosensor’s performances are affected by the amount of immobilized ALP, surface area of the electrode and method of enzyme immobilization. In this research, alkaline phosphatase was immobilized on chitosan membrane modified by glutaraldehyde which was coated on the surface of SPCE. Diazinon measurements were carried out at pH 8.5, using 0.325; 0.650; 0.975; 1.30; 1.65 mg alkaline phosphatase. The results showed that optimum performance is obtained at 1.65 mg ALP immobilized by crosslink, with a range of concentrations (0.27 to 1.5) ppm, 32.61 mV/ppm sensitivity, 0.27 ppm of LOD and it can be used to measure 6 samples diazinon.
