Abstract

Tirosin kinase merupakan enzim yang mengatur hubungan antar sel, diferensiasi, adhesi serta pengaturan sel. Aktivitas tirosin kinase penting dalam proses autofosforilasi spermatozoa dan aktivitasnya akan maksimal bila bekerja pada kondisi optimum. Tirosin kinase diisolasi dari spermotozoa tikus (Rattus norvegicus) jantan dengan cara sentrifugasi dan ultrasonikasi. Pada penelitian ini ditentukan kondisi optimum tirosin kinase dengan variasi pH (6; 6,5; 7; 7,5 dan 8), temperatur (20; 25; 30; 35 dan 40)0C, waktu inkubasi (20; 25; 30; 35; dan 40) menit, dan untuk penentuan berat molekul digunakan metode SDS-PAGE. Hasil dari penelitian ini menunjukkan bahwa kondisi optimum aktivitas tirosin kinase hasil isolasi spermatozoa tikus (Rattus norvegicus) jantan pada pH 7, temperatur 350C, waktu inkubasi 30 menit dengan aktivitas 0,263 unit dan didapatkan berat molekul sebesar 94,61 kDa. Hasil uji statistik menunjukkan bahwa pH, temperature, dan waktu inkubasi memberikan pengaruh sangat nyata (P<0,01).

English Abstract

Tyrosine kinase is an enzyme which regulates the relationships between cell adhesion, differentiation, and cell settings. Tyrosine kinase activity is important in the process of autofosforilasi sperm and its activities will be maximum when the working conditions of optimum. Tyrosine kinase isolated from spermotozoa rats (Rattus norvegicus) males by centrifugation and ultrasonikasi technique. In this research specified conditions optimum tyrosine kinase with variations of pH ( 6; 6,5; 7; 7.5 and 8), temperature ( 20; 25; 30; 35 and 40 )0C, incubation periods ( 20; 25; 30; 35; and 40 ) minutes, and for the determination of molecular weight used method of SDS-PAGE. The result of this research showed that the optimum activity tyrosine kinase from sperm of rat (Rattus norvegucus) male at pH 7, temperature 350C, and 30 minutes incubation time with 0,263 unit activity and obtained by molecular weight 94,61 kDa. The result of statistic analyzed showed the variation of pH, temperature, and incubation time give an very obvious influence (P<0,01).
