Abstract

Telah dilakukan penelitian untuk menentukan kondisi optimum urease yang diamobilisasi dalam kitosan-natrium tripolifosfat. Urease yang digunakan diisolasi dari Schizzosaccharomyces pombe dan dimurnikan melalui metode pemurnian bertingkat menggunakan amonium sulfat fraksi 30–45%. Urease merupakan enzim yang memiliki kemampuan untuk menghidrolisis urea menjadi ammonia dan karbondioksida. Aktivitas urease ditentukan berdasarkan jumlah mikromol ammonia yang dihasilkan per mL enzim per menit dimana kadar ammonia ditentukan dengan spektrofotometri melalui penambahan reagen Nessler. Urease diamobilisasi menggunakan metode penjebakan dalam kitosan-natrium tripolifosfat. Kondisi optimum amobilisasi urease dipelajari dengan menentukan aktivitas tertinggi pada variasi konsentrasi kitosan (1,5 ; 2,0 ; 2,5 ; 3,0 ; 3,5) % w/v, konsentrasi urease (0,840 ; 1,680 ; 2,520 ; 3,360 ; 4,200) mg/mL, dan efisiensi penggunaan ulang urease amobil. Hasil penelitian menunjukkan bahwa konsentrasi kitosan optimum yaitu 2,5% (w/v) dan konsentrasi urease optimum pada 2,520 mg/mL dengan aktivitas tertinggi sebesar 0,57 unit. Efisiensi penggunaan urease amobil menggunakan matrik kitosan-natrium tripolifosfat dapat digunakan sebanyak 4 kali dengan aktivitas sebesar 65,62%.

English Abstract

The research to determine optimum condition of immobilized urease has been done. Urease was isolated from Schizzosaccharomyces pombe and was purified through fractional purification method using ammonium sulphate saturation of 30– 45%. Urease is an enzyme that has the ability to hydrolyze of urea into carbon dioxide and ammonia. The activity of urease was defined as the amount of micromol ammonia per mL enzyme per minute and resulted ammonia was analyzed by spectrofotometri using Nessler reagen. Urease was immobilized by entrapment method in chitosan-sodium tripolyphosphate. The optimum condition of immobilization urease was studied by determining the highest activity in various chitosan concentration (1.5 ; 2.0 ; 2.5 ; 3.0 ; 3.5) % w/v, urease concentration (0.840 ; 1.680 ; 2.520 ; 3.360 ; 4.200) mg/mL, and efficiency of immobilized urease. The result of this experiment showed that the optimum chitosan concentration was 2.5% w/v while the optimum urease concentration was 2.520 mg/mL with the highest activity of 0.57 units. Efficiency of immobilized urease using chitosan-sodium tripolyphosphate matrix can be performed four repetitions with activity of 65.62%
