Abstract
Penelitian ini bertujuan untuk mengetahui pengaruh pemberian puerarin terhadap penurunan ekspresi VCAM-1 pada kultur sel endotel (HUVECs) yang diinduksi leptin. Kultur sel endotel dikelompokkan menjadi dua, yaitu (1) perlakuan puerarin (0, 5, 25, 200 dan 525 μM) selama 6 jam dan (2) leptin (25 ng/mL) selama 6 jam yang diikuti dengan pemberian puerarin (0, 5, 25, 200 dan 525 μM) selama 6 jam berikutnya. Selanjutnya dilakukan pewarnaan imunositokimia menggunakan mouse monoclonal VCAM-1 sebagai antibodi primer dan anti mouse VCAM-1 sebagai antibodi sekunder. Data yang diperoleh dianalisis menggunakan one way Anova dan dilanjutkan dengan uji Tukey . Hasil penelitian menunjukkan bahwa leptin 25 ng/mL mampu meningkatkan ekspresi VCAM-1 (2,68 ± 0,15) % dibandingkan dengan leptin 0 ng/mL (0,54 ± 0,15) %. Namun pemberian puerarin (5, 25, 200 dan 525 μM) tidak signifikan dalam menurunkan ekspresi VCAM-1.
English Abstract
The aim of this research was to determine the influence of puerarin to decrease the VCAM-1 expression in HUVECs culture that was exposed by leptin. The endothelial cell cultures were divided into two groups that were (1) induced by puerarin (0, 5, 25, 200, 525 μM) for 6 hours and (2) induced by leptin (25 ng/mL) for 6 hours, then incubated with puerarin (0, 5, 25, 200, 525 μM) for another 6 hours. The imunocytochemistry was employed by mouse monoclonal antigen antibody VCAM-1 as primer antibody and anti mouse VCAM-1 as secondary antibody. Data were analyzed by using one way Anova and followed by Tukey test. The result showed that leptin 25 ng/mL could increase the VCAM-1 expression (2,68 ± 0,15) % compared to leptin 0 ng/mL (0,54 ± 0,15) %. However the treatment of several doses of puerarin (5, 25, 200, 525 μM) were did not affect the VCAM-1 expression significantly. Key words: HUVECs, leptin, puerarin, VCAM-1.
