Abstract
Ekstraksi ampas teh hitam menghasilkan polifenol yang tinggi dengan menggunakan metode MAE (Microwave Assisted Extraction) dan pelarut metanol 60%. Tujuan dari penelitian ini adalah untuk optimalisasi faktor lama pemanasan pada MAE, dan konsentrasi alkohol terhadap respon rendemen, antioksidan, dam kadar katekin. Metode Microwave Assisted Extraction (MAE) merupakan teknik ekstraksi relatif baru yang mengkombinasikan energi gelombang mikro dan teknik konvensioal dengan pelarut. Pergerakan yang sangat cepat ini menghasilkan friksi atau gesekan dan akhirnya menghasilkan energi panas dalam bahan sehingga dinding sel maupun jaringan bahan akan rusak, dan solute akhirnya dapat keluar. Hasil penelitian menunjukkan perlakuan penggunaan konsentrasi pelarut alkohol dengan konsentrasi 62,74% dan lama pemanasan 8 menit yang mempunyai ketepatan (desirability) sebesar 74,20%. Perlakuan optimal tersebut menghasilkan prediksi rendemen 54,8399%, kandungan antioksidan (IC50) 7,50516 ppm dan kadar katekin sebesar 92,0558%. Hasil verifikasi diperoleh nilai rendemen 54,62%%, kandungan antioksidan (IC50) 14,38847, dan kadar katekin sebesar 94,74% dengan simpangan masing-masing respon berturut-turut 0,403%, 47,43%, dan 2,833%.
English Abstract
Black tea waste extraction has the highest polifenol extraction using MAE (Microwave Assisted Extraction) and 60% of metanol solvent. The purpose of this research was to optimize factors of heating time and alcohol solvent for yield response, antioxidant (IC50), and catechin content. MAE (Microwave Assisted Extraction) is the method of extraction that combined microwave energy and conventional method with an organic solvent. The excess of this method can get the result of material extraction in a minuts, the microwave energy gives molecular movement with ion migration and dipol rotation. The fast movement will result friction and then will produce the heat energy in materal, so the membran cell or material’s tissue will damage , after that solute will turn out from the material. Optimum result from the alcohol solvent treatment was using 62,74% and the time to heating the material was 8 minuts, and the accuracy (desirability) about 74,20. The optimal treatment resulted in yield prediction 54,8399%, Antiocsidant content (IC50) 7,50516 ppm, and level of catechin is about 92,0558%. Verification results obtained resulted the yield was about 54,62%, Antiocsidant content (IC50) was about 14,38847 ppm, and level of catechin was about 94,74%, with deviation of each responses respectively 0,403%, 47,43%, and 2,833%
