Abstract

Diabetes Mellitus tipe 2 (DMT2) adalah gangguan metabolisme yang ditandai dengan adanya resistensi insulin dan penurunan fungsi sel pankreas. DMT2 yang berlangsung lama dapat menyebabkan komplikasi ke berbagai organ akibat keadaan hiperglikemia yang muncul dan peningkatan produksi radikal bebas. Latihan fisik diharapkan dapat mengurangi kerusakan dan gangguan pada jaringan tubuh dengan meningkatkan metabolisme glukosa dan sensitivitas insulin. Penelitian ini bertujuan untuk mengetahui pengaruh latihan fisik (treadmill) terhadap kadar Malondialdehyde (MDA) jantung dan gambaran histopatologi jantung pada tikus (Rattus norvegicus) model DMT2. Tikus model DMT2 dihasilkan dengan induksi 40 gram/ekor/hari pakan tinggi lemak (High Fat-Diet) selama 63 hari dan multiple low-dose Streptozotocin 30 mg/kg secara intraperitoneal. Penelitian ini menggunakan tiga kelompok tikus, yaitu kelompok normal sedentari, DMT2 sedentari dan DMT2 latihan fisik. Latihan fisik diberikan 6 hari dalam seminggu selama 10 minggu, dengan kecepatan dan durasi yang meningkat setiap 2 minggu, dimulai dari 10 meter/menit selama 10 menit hingga mencapai 27 meter/menit selama 60 menit. Kadar MDA diukur dengan uji Thiobarbituric Acid Reactive Substance (TBARS) secara spektrofotometri dan gambaran histopatologi jantung diwarnai menggunakan Hematoxylin-Eosin (HE). Hasil penelitian menunjukkan bahwa latihan fisik (treadmill) dapat menurunkan kadar MDA jantung secara signifikan (p<0.05) dan memperbaiki kerusakan myocardium yang terlihat dari berkurangnya deformasi nukleus dan myofibril disintegrasi, sehingga dapat disimpulkan latihan fisik (treadmill) dapat menurunkan kadar MDA jantung dan memperbaiki histopatologi jantung pada tikus model DMT2.

English Abstract

Type 2 Diabetes Mellitus (T2DM) is a metabolic disorder marked by insulin resistance and decreased function of pancreatic beta cell. Long-standing T2DM can lead to various organ complications due to hyperglycemia and increased production of Reactive Oxygen Species (ROS). Physical exercise is expected to reduce the damage and disruption in the body's tissues by increasing glucose metabolism and insulin sensitivity. This study was aimed to determine the effects of physical exercise (treadmill) on cardiac malondialdehyde (MDA) level and cardiac histopathology in rat (Rattus norvegicus) model of T2DM. Rat model of T2DM was created from induction of 40 g/head/day of high-fat feed for 63 days and multiple low-dose streptozotocin 30 mg/kg intraperitoneally. Rats were divided into three groups consist of normal sedentary, T2DM sedentary and T2DM physical exercise. Physical exercise is given 6 days a week for 10 weeks, with speed and duration increased every 2 weeks, starting from 10 meters/min for 10 minutes to 27 meters/minute for 60 minutes. MDA levels were measured by Thiobarbituric Acid Reactive Substance (TBARS) assay and cardiac histopathology stained using Hematoxylin-Eosin (HE). The results showed that physical exercise (treadmill) can significantly reduce cardiac MDA levels (p<0.05) and repair myocardium damage seen from reduced deformation of the nucleus and myofibril disintegration, so it can be concluded that physical exercise (treadmill) can reduce cardiac MDA levels and improve cardiac histopathology in rat model of T2DM.
