Abstract
Asap rokok mengandung radikal bebas eksogen yang tergolong dalam Radical Oxygen Species (ROS) yang akan meningkatkan akumulasi radikal bebas di dalam tubuh. Sehingga menyebabkan ketidakseimbangan antara oksidan dan radikal bebas di dalam tubuh. Ketidakseimbangan ini akan mengakibatkan stress oksidatif yang dapat meningkatkan aktivitas protease dan memicu peroksidasi lipid yang merusak jaringan. Radikal bebas ini dapat dihambat dengan antioksidan eksogen yang berupa Thymoquinone yaitu sisi aktif pada Jintan Hitam (Nigella sativa L.). Tujuan penelitian ini untuk mengetahui pengaruh ekstrak jintan hitam (Nigella sativa L.) terhadap pencegahan peningkatan aktivitas protease dan kerusakan gambaran histopatologi trakea tikus putih (Rattus norvegicus) yang dipapar asap rokok. Penelitian ini menggunakan hewan coba tikus putih (Rattus norvegicus) jantan berumur 8-12 minggu dengan berat badan 150-200 gram. Penelitian ini terdiri dari kelompok kontrol negatif (tikus putih sehat), kontrol positif yang dipapar asap rokok sebanyak 2 batang/kelompok/hari setiap pagi dan sore selama 21 hari dan kelompok perlakuan yang diberi ekstrak jintan hitam (Nigella sativa L.) dengan dosis masing-masing kelompok 0,6g/kgBB/hari, 1,2g/kgBB/hari dan 2,4g/kgBB/hari kemudian diberi paparan asap rokok 2 batang/kelompok/hari setiap pagi dan sore selama 21 hari. Hasil penelitian menunjukkan bahwa pemberian ekstrak jintan hitam (Nigella sativa L.) dosis 2,4g/kgBB/hari mampu mencegah peningkatan aktivitas protease sangat signifikan (P<0,01) hingga mencapai penurunan 78,30% dan menghambat kerusakan histopatologi epitel, silia, sel goblet dan sekresi mukus trakea tikus putih (Rattus norvegicus) yang dipapar asap rokok. Kesimpulan dari penelitian ini adalah ekstrak Jintan Hitam (Nigella sativa L.) dapat mencegah peningkatan aktivitas protease dan kerusakan histopatologi trakea.
English Abstract
Cigarette smoke contains exogenous free radicals belonging to the Radical Oxygen Species (ROS), which would increase the levels of free radicals in the body. This creates an imbalance between oxidants and free radicals in the body. This imbalance will increase protease activity and lipid peroxidation that can damage tissues. These free radicals can be inhibited with exogenous antioxidants such as Thymoquinone which is an active compound in Black Cumin (Nigella sativa L.). The purpose of this study is to determine the effect of Black Cumin (Nigella sativa L.) extract to prevent an increase in protease activity and damages in histopathological picture of white rats’ trachea that exposed to cigarette smoke. This study used white male rats (Rattus norvegicus) aged 8-12 weeks with body weight 150-200 grams. This research used five group of rats that divided into negative control (healthy rats) was given black cumin (Nigella sativa L.) extract, positive control induced by cigarette smoke 2 cigarette/group/day every morning and evening for 21 days without preventive treatment of black cumin extract (Nigella sativa L.), and three treatment groups was given black cumin with 0,6g/KgBW/day,1,2g/KgBW/day, 2,4g/KgBW/day doses of each group and then induced by cigarette smoke 2 cigarette/group/day every morning and evening for 21 day. The results observed in the form of protease activity and histopathological picture trachea.The results showed that rat group with Black Cumin extract administration with 2.4g / KgBB / day dose was able to prevent the increase of protease activity significantly (P <0.01) up to 78.30% and inhibit the histopathology of epithelium, cilium, goblet cell and mucus of rats (Rattus norvegicus trachea exposed to cigarette smoke. The conclusion of this study is black cumin extract (Nigella sativa L.) can prevent the increase of protease activity and histopathologic damage of trachea.
